Objective: This study observed brain IL-15 content changes in aging PD mice and the effect of prior-exercise training on the content of brain IL-15 in PD mice,analyzed the relationship between IL-15 induced by prior-exercise and autophagy,apoptosis and mitochondrial function,explored the neuroprotective role and mechanism of IL-15 in Parkinson's disease.Methods: 1?90 Male C57BL/6 mice were divided into young,middle and old control groups(YC,MC,OC),young,middle and old PD groups(YM,MM,OM).Behavior changes of all groups were observed;HE dyeing to observe brain tissue changes;IL-15 content of brain tissue,skeletal muscle tissue and serum were measured by ELISA method;brain mitochondrial function index were detected.2?90 Male C57BL/6 mice were divided into control group,endurance and resistance exercise training groups(C,E,R),PD group,the prior endurance and resistance exercise training PD groups(MC,ME,MR).Based on the test content of first part,brain tissue IL-15 mRNA,autophagy and apoptosis related gene expression were determined by RT-PCR and Western blot;Brain cell apoptosis were detected by TUNEL method.3?wild(WT)and skeletal muscle(MKO)IL-15-/-mice were divided into control groups(C)and acute exercise groups(E).IL-15 content of brain tissue,skeletal muscle and serum were measured by ELISA method.Results: 1?Brain mitochondrial function of OC group and OM group reduced significantly,IL-15 content and IL-15 mRNA expression in brain tissue increased significantly.Brain tissue IL-15 content and mitochondrial function indexes correlated negatively.2?Brain mitochondrial function of ME group and MR group increased significantly;IL-15 content in brain tissue increased significantly;autophagy gene expression increased significantly;pro-apoptosis gene expression of ME and MR groups were significantly reduced;except the BCL-2 in MR group,anti-apoptosis gene expression in two groups were significantly increased;brain IL-15 content of PD mice was positively correlated with autophagy and anti-apoptosis gene expression,was negatively correlated with the pro-apoptosis gene and apoptosis index,were correlated positively with mitochondrial function indexes.3?Compared with WT-C group,IL-15 content of brain tissue,skeletal muscle tissue and serum in WT-E group increased significantly;Compared with MKO-C group,IL-15 content of brain tissue,skeletal muscle tissue and serum in MKO-E group did not change significantly.Conclusion:1?IL-15 content of brain tissue in aging mice and PD mice increased,which may be a compensatory response against the decline of brain mitochondrial function.2?Endurance and resistance exercise training all can increase IL-15 content of brain tissue in PD mice,the prior-exercise training induced IL-15 take part in the process,that exercise increased autophagy and inhibited apoptosis,which improved mitochondrial function to exert neuroprotective role in PD.3?Exercise-induced IL-15 may be muscle derived.
