were used to ignite the ePilePtic model.Finally,the rats were randomly assigned to the following groups:model group,low,middle and high dose of 10 groups,and low,medium and high dose group of 21 days.The general state and behavioral changes of the rats in the corresPonding group were observed at the first 1 days of treatment and the course of treatment for tenth days and 21 days.The Pathological changes of hiPocamPus were observed by HE staining.The exPression of Bax,Bcl-2 and c-fos PKCa,c-jun Protein and IL-1β,IL-6 were detected by immunohistochemistry.The final experimental data were inPut SPSS 17.0 statistical software for statistical Processing,P<0.05 was significant difference,(P<0.01)was very significant difference.ResultTo cycle model,remove the death and did not reach the state of the kindling rats,a total of the remaining 70,70 rats were randomly assigned to model group,Shi Gan Powder is low,medium and high dose group of 10 days and low in the high dose group of 21 days.1.general state and body mass changeThe general state of the model group was worse than that of the blank group,and the number of each group was better than that of the model group,the middle and high dose group was better than the low dose group,and the 21 day group was better than the group of 10 days.The changes of body weight,before and after the treatment of gastric lavage in the model group and the group has a significant or significant difference(P<0.05 or P<0.01).The middle and high dose group had significant difference comPared with the model group(P<0.05).Low,medium and high dose group by gavage for 10 body weight were significant or extremely significant increase(P<0.05 or P<0.01).After 21 days of treatment,the quality of each group was significantly higher than that of model group(P<0.05).In the high dose group gavage after treatment significantly increased(P<0.01).There was significant difference in body mass between low and high dose groups(P<0.05).2.behavioral observationNo seizures in the blank group.In the 1 days before treatment,the level of ePilePtic seizures in all groups were in the level of grade 4 or above,after treatment for about 10 days,comPared with the model group,the middle and high dose group had significant change(P<0.05).Low dose group had a significant decrease,but the results were not significantly difference between the low and high dose groups(P<0.01).For the latency of seizures,in different groups by gavage for 10 days and 21 days,seizure latency were significantly Prolonged in the high dose hiPPocamPal cells in the blank group were normal.The number of cells in the model group was significantly reduced,the level of disorder,swelling and deformation of cells,cytoPlasmic staining deePened,Pyknosis,fragmentation.ComPared with the model group,the imProvement of the middle and high dose group was higher than that of the low dose group,and the 21 day group was greater than the 10 day group.4.ExPression ofc-fosThe number of c-fos Positive cells increased significantly in the model group.There was a decrease in all groups.There was no significant difference between the 21 groups excePt for the significant difference between the low dose and high dose(P<0.05)in the 10 day group and the second day group.In addition,there was a significant or very significant reduction in(P<0.01 or P<0.05)in each group comPared with the model group.The low dose group and high dose group were significantly lower(P<0.05)in the same dose 10 day group and the 21 day group,and the decrease rate of the 21 day group was higher than that of the 10 day group.5.Expression of baxIn the treatment of 10 days and 21 days,low and high dose group,the results were significantly different(P<0.05),middle and high dose group and model group was significantly lower(P<0.05).However,the middle and high dose 10 days group and the 21 day group had significant or extremely significant reduction(P<0.05 or P<0.01),but the low dose group was significantly decreased,but the results were not statistically significant(P>0.05).6.ExPression ofBcl-2In each dose group of 10 days and 21 days group,low and high high dose group was significantly higher(P<0.05),and the 21 day group was significantly larger than the 10 day group.In the middle and high dose group,there was significant or very significant increase in(P<0.05 or P<0.01).8.ExPression of c-junThere were significant differences between the 10 groups in the treatment group and the control group in the 21 day group,and there was significant difference between the low and high dose groups(P<0.05).ComPared with the model group,the middle and high dose group had significant difference(P<0.05),but the 21 day group and the high dose group changed significantly.There was significant or very significant difference between the 10-day and the high-dose group(P<0.05 or P<0.01)in the middle and high dose group(P<0.05 or P<0.01).9.Expression of IL-1βThere was significant difference between the 10 and the 21 days in the group of low and high dose of(P<0.05).ComPared with the model group,the middle and high dose group was significantly lower(P<0.01),and the changes were more obvious in the 21 day group and the high dose group.There was significant or very significant difference between the 10-day and the high-dose group(P<0.05 or P<0.01)in the middle and high dose group(P<0.05 or P<0.01).10.Expression of IL-6There were significant or extremely significant differences between the 10 and the 21 day groups in the low and high dose groups(P<0.05 or P<0.01),respectively.ComPared with the model group,the middle and high dose group had significant difference(P<0.01),but the 21 day group and the high dose group changed more significantly.There was significant or very significant difference between the 10-day and the high-dose group(P<0.05 or P<0.01)in the middle and high dose group(P<0.05orP<0.01).Conclusion1 stone
