Objective:to study the Wen Xin Dan on myocardial ischemia reperfusion injury model of rat myocardial protection and related inflammatory mechanisms.Methods:1.Different Wen Xin Dan compatibility proportion group,control group,model group,pretreatment,respectively effects on myocardial ischemia reperfusion injury model in rats,to detect the rat serum CK-MB,cTnI levels,the influence of the area of myocardial infarction(mi);Wen Xin Dan compatibility proportion relation examination,optimizing the best compatibility proportion metacentric Dan group.2.Using the tem observation of Wen Xin Dan to myocardial ischemia reperfusion injury model of rat myocardial ultra micro structure,the influence of immunohistochemical method to detect Wen Xin Dan T-Akt in myocardial tissue,the expression of p-Akt.To explore Wen Xin Dan mechanism against myocardial ischemia-reperfusion injury.3.By western blot method and immunohistochemical method to detect Wen Xin Dan on myocardial ischemia reperfusion injury model of rat myocardial tissue in the nf-kappa B,the influence of protein distribution and expression of cox-2.4.Observe the Wen Xin Dan to myocardial ischemia reperfusion injury model in rats serum TNF alpha,beta,IL-6,IL-1 the effect of IL-10 levels,explore its anti-inflammatory action mechanism.Results:1.Wen Xin Dan each treatment group can significantly reduce the myocardial ischemia reperfusion injury model of rat serum CK-MB,cTnI levels,reduce the model of rat myocardial infarction area,among which effect the most significant Wen Xin Dan 1:2:1.2.Wen Xin Dan 1:2:1 group compared with model group can significantly improve the myocardial ischemia reperfusion injury model of rat myocardial ultra micro structure;Improve the myocardial tissue of p-Akt protein expression.3.The Wen Xin Dan 1:2:1 group compared with model group can significantly inhibit myocardial ischemia-reperfusion injury model of rat myocardial tissue in the nf-kappa B,protein distribution and expression of cox-2.4.Wen Xin Dan 1:2:1 group compared with model group can significantly reduce the myocardial ischemia reperfusion injury model in rats serum TNF alpha,beta,IL-6,IL-1 content,increase the content of IL-10.Conclusion:1.Different Wen Xin Dan compatibility proportion group can significantly reduce the myocardial ischemia-reperfusion injury model of rat myocardial enzyme CK-MB,cTnI levels,reducing myocardial infarction area,optimizing the Wen Xin Dan 1:2:1 group for against myocardial ischemia reperfusion group with the best compatibility proportion.2.Wen Xin Dan through the activation of PI3K/Akt signal transduction pathway,improve the myocardial tissue of p-Akt protein expression,reduce the nf-kappa B,cox-2 protein expression,reduce myocardial tissue damage degree,give play to the role of myocardial protection.3.Wen Xin Dan through the activation of PI3K/Akt signal transduction pathway,cut inflammation factor TNF alpha,beta,expression of IL-6,IL-1 and IL-10 can promote inflammation suppression factor release,thus play a role of myocardial protection.
