Objective: 1.To estimate the relationship between the risk of preeclampsia and two thrombophilia gene single-nucleotidepolymorphisms(SNPs),the factor V G1691 A SNP and the prothrombin G20210 A SNP.2.To study the difference and distribution of the factor V G1691 A SNP and the prothrombin G20210 A SNP genotypes and allele frequency in Xinjiang Kazak population and Han population with preeclampsia disease(PE).3.To study the coagulation gene,platelet parameter and the related research of preeclampsia in Xinjiang Kazak population and Han population with preeclampsia disease(PE).Method: 1.Date Sources A systematic search of the English-language literature up to 2014 was performed using Medlineand EMBASE,Elsevier science Search terms included “preeclampsia,” “thrombophilia,” “factor V Leiden,” “prothrombin gene 20210,” and their combinations.2.PE patients were collected from January 2013 to December 2015 at the first Affiliated Hospital of Xinjiang Medical University,hospitalized 100 cases,including 50 cases of Kazak and Han 50 cases;Kazak healthy control group of 50 patients,50 cases of Han nationality.Polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP)technique to detect FVL and PT G20210 A gene G1691 A gene polymorphism association analysis FVL and PT G20210 A gene G1691 A gene polymorphism and the PE.Logistic analysis to study coagulation gene,platelet parameter and related research of preeclampsia in Xin Jiang Kasak and Han nationality.Result 1.Thirty-seven studies with 5048 preeclampsia patients and 6796 controls were included in the meta-analysis.We found that the prothrombin G20210 A polymorphism was associated with an increased risk of all preeclampsia(pooled odds ratio(OR)=1.81,95% confidence interval(CI)1.25-2.63)and severe preeclampsia(pooled OR=3.02,95%CI 2.06-4.45).Meanwhile,the pooled OR for the association between factor V Leiden and all preeclampsia was 1.60(95%CI 1.28-2.00)and 2.45(95%CI 1.63-3.69)for the cases of severe preeclampsia.2.PT gene G20210 A three genotype in Kazakh PE patients and the control group were: GG type [36%(18/50)ratio of 22%(11/50)],GA type [44%(22/50)ratio of 54%(27/50)] and AA-type [20%(10/50)ratio of 24%(12/50)];patients and controls the distribution of gene frequencies Han PE were: GG type [30%(15/50)ratio of 16%(8/50)],GA type [52%(26/50)ratio of 62%(31/50)] and AA-type [18%(9/50)ratio of 22%(11/50)],FVL gene G1691 A three genotype in Kazakh PE patients and the control group were: GG type [40%(20/50)ratio of 26%(13/50)],GA type [48%(24/50)ratio of 54%(27/50)] and AA-type [12%(6/50)than 14%(7/50)];Han patients and the control group gene frequencies of PE are: GG type [34 %(17/50)ratio of 28%(14/50)],GA type [50%(25/50)ratio of 58%(29/50)] and AA-type [16%(8/50)than 14%(7/50)],PT gene single nucleotide polymorphism(SNP)GG genotype in comparison with genotype national cases and controls,the difference was statistically significant(Kazakh: X2=6.069,P=0.048;Han X2=6.181,P=0.045);and,in the comparison between the two peoples and PE control group,the difference was statistically significant(P<0.05).FVL gene single nucleotide polymorphism(SNP)GG genotype frequencies genotype compared with national case and control group was statistically significant difference(Kazakh: X2=5.018,P=0.029;HanX2=6.67,P=0.025);and in the comparison between the two peoples and PE control group,the difference was statistically significant(P < 0.05).3.TT(P=0.012,OR=1.386,95%CI: 0.184-0.813)?APTT(P=0.029,OR=1.087,95%CI: 1.035-1.141)?D-Dimter(P=0.031)are risk fators in Xinjiang Kazak population with preeclampsia disease(PE),PT(P=0.133)?FIB(P=0.211)?FDP(P=0.327)?PCT((P=0.668);there is no relationship in FIB,FDP,PCT and Kasak people with preeclampsia disease.PLT(P=0.031,OR=1.699,95%CI: 1.181-2.743);MPV(P=0.001,OR=1.931,95%CI: 1.717-2.426);PDW(P=0.001,OR=2.679,95%CI: 2.112-3.142)are risk fators in Xinjiang Kazak population with preeclampsia disease(PE).Conclusion.1.This meta-analysis supports that the factor V G1691 A SNP and the prothrombin G20210 A SNP are associated with an increased risk for both preeclampsia overall and severe preeclampsia.2.Prothrombin(PT)gene G20210 A and coagulation factor V(FVL)gene expression levels G1691 A in preeclampsia placenta significantly increased,and the Xinjiang Kazakh expression levels were significantly higher than the Han.3.TT,APTT?D-Dimter are risk fators in Xin jiang Kazak population with preeclampsia disease.there is no relationship in FIB,FDP,PCT and Kasak people with preeclampsia disease.PLT;MPV;PDW are risk fators in Xinjiang Kazak population with preeclampsia disease(PE).
