Mapping the connection of brain networks help us understand the function of the brain.It's a must that developing more and more technology for brain mapping.Traditional methods for neural circuits tracing depend on Golgi staining?protein or dye.Virus-based tracing methods which were developed quickly have been widely used in neural circuits research recently.Neurotropic viruses used in neural circuits contain pseudorabies virus(PRV),rabies virus(RV),vesicular stomatitis virus(VSV),herpes simplex virus(HSV).These virus family could be applied in mapping the monosynaptic or multi-synaptic input and output of certain neurons.Otherwise,another virus used in manipulating neural circuits are adeno-associated virus(AAV).A lot of questions in neuroscience could be solved by these virus tools.The research of this paper focus on following aspects:1?Do something in founding and improving the virus system for mapping neural circuits in our lab.2?Co-research the neural circuit of the opposite monosynaptic scaling of BLP-vCAl inputs governs hopefulness-and helplessness-modulated spatial learning and memory.Our study demonstrates the change of spatial learning and memory under different emotional states.The results indicated that monosynaptic scaling of posterior baso-lateral amygdala(BLP)-ventral hippocampal CA1(vCA1)controls emotion-modulated spatial memory.This project was cooperated with Prof.Jianzhi Wang of Huazhong University of Science and Technology(HUST).3?Co-research the new technology of sparse labeling combined with fMOST to trace the pattern of single neuron projections in the whole-brain.This method was accomplished by employing a special designed adeno-associated virus(AAV).We demonstrate the data by injecting the special AAV into DLO which was a sub-region of mPFC and reconstructing the single cell full morphology of DLO.This new method may help scientists understand fine projection information of single neuron.This project was collaborated with Prof.Yuhui Zhang and Prof.Shaoqun Zeng of HUST.4?Co-research the fine morphology of mossy cell in dentate gyrus by injecting AAV Sparse virus into dentate gyrus of C57BL/6 mouse.This virus could specifically label the mossy cell in high brightness.Also,single mossy cell morphology could be reconstruct by combing the fMOST method and single cell tracing method.This project was collaborated with Prof.Yuhui Zhang and Prof.Shaoqun Zeng of HUST.
