Objective The aim of this article is to describe a novel nanocomposite gel fiber encapsulated human placental extract through layer-by-layer self-assembly, and explore the treatment effects of chronic ulcer in diabetic rats with self assembling nanofiber gel encapsulated-polydeoxyribonucleotide.Methods The 10 mg of IKVAV, RGD and FGL-PA were screened and combined to produce an optimal vehicle nanofiber gel through layer-by-layer assembly. We detected its morphology, particle size, drug loading efficacy, the encapsulation rate, release efficiency and the structure validation after it was diluted 0,50,100 times. Diabetic skin ulcer mouse model was established. They were divided into control group, common wound group and infectious wound group. Human embryonic fibroblast (H2EL) cells and vascular (?)dothelial (VE) cells were treated with short poly-N-acetyl glucosamine nanofibers and polydeoxyribonucleotide. Their effects on cell proliferation, revascularization and inhibiting infection were detected by RT-PCR, Western-bloting, HE staining and immunohistochemical methods.Results The optimal proportion and concentration of nanometer fiber gel: IKVAV:RGD:FGL-PA=1:1:1,0.6mg/ml by observing the liquidity, diffusion time and viscosity. The nanometer fiber gel had slow-releasing profile; its average particle size of 212.8 nm, and the slow release time was more than 4 days. The preliminary results confirmed that the structure of HPE encapsulated nanofiber gel was unvaried compared with the HPE structure. The expression levels of cytokines and angiogenic factors increased in the treatment groups especially in sNAG encapsulated-PDRN group. HE staining results indicated that PDRN, sNAG and sNAG encapsulated-PDRN could improve the wound healing, immunohistochemical results showed that PDRN, sNAG and sNAG encapsulated-PDRN promoted cell proliferation and new vessel formation especially sNAG encapsulated-PDRN.Conclusions This kind of novel composite nanocapsules may offer a promising delivery system for human placenta extract. sNAG encapsulated-PDRN may have a potential application in the treatment of diabetic ulcers and chronic wound healing.
