Moso bamboo?Phyllostachys edulis?,is the most important bamboo species in China and is widely used in timber production,ecological construction,and cultural exchange.It plays an important role in addressing environmental degradation and relieving shortage of resources.Moso bamboo is characterized by infrequent sexual reproduction.In addition,they often synchronously flower,and collectively die after flowering.Here,in order to discovery the floral development mechanism of moso bamboo,the morphogenesis of indefinite inflorescence of moso bamboo was studied by paraffin section techniques;transcriptome sequencing and four expression profiles with the RNA-Seq sequencing technology of moso bamboo flowers were studied;and the function of PheDof-1 and PheMADS-14 was researched by qRT-PCR,western blot and in situ hybridization.The major findings are as follows:1.Moso bamboo flowers are fake inflorescences and spicate inflorescences,average length of the inflorescences is about 7-9 cm,an inflorescence has several fake spikelets,and each spikelet has 1-6 florets.A floret has 2 glumes,1 palea,1 lemma,3 lodicules,3 stamens and 1 pistil.The morphogenesis of inflorescence could be divided into four periods:floral bud formation stage,inflorescence growing stage,bloom stage,and embryo formation stage.2.Ttranscriptome sequencing with Illumina HiSeqTM 2000 sequsncing technology for moso bamboo was performed,we obtained 5 G raw data,68,305,136 and 67,617,866 clean reads,respectively.By comparing the two libraries at a statistically significant value?FDR?0.001 and log2Ratio?1?,11,703 genes were detected changing expression,3,921 and3,301 in which the genes involved in RNA transport and mRNA surveillance pathway,1,582genes involved in plant-pathogen interaction pathways and 1,293 genes involved in plant hormone signal transduction pathways.3.The F1,F2,F3,F4 expression profiles obtained more than 10 million clean reads,respectively.With the statistics analysis of the differential expression genes,we found that there were 9,735 genes up-regulated and 2,859 genes down-regulated in the CK/F1 genes;there were 10,055 genes up-regulated and 2,874 genes down-regulated in the CK/F2;there were 9,237 genes up-regulated and 3,096 genes down-regulated in the CK/F3;there were 6,172genes up-regulated,3,615 genes down-regulated in the CK/F4,respectively.4.When clustering DEGs into different groups,we found that the two major groups,the up-regulated group and the down-regulated group,showing different enrichments of DEGs:plant hormone signal transduction,protein processing in endoplasmic reticulum and plant-pathogen interaction in the up-regulated group as opposed to photosynthesis,photosynthesis-antenna proteins and carbon fixation in photosynthetic organisms in the down-regulated group.5.By applying Illumina Hi SeqTM 2000 sequsncing technology on studying transcriptom and differentially expressed genes profiles in moso bamboo,we found a number of transcription factor families with stress,including CMLs,WRKY,NAC,RAD23,HSP20/70/90,Dof,JAZ,MYC2,FLS2,and PR1/RPS2/RPS5.6.PheDof-1 was grouped into MCOG C Cluster,and PheMADS-14 was grouped into AP/AGL9 Cluster.Through the in situ hybridization,PheDof-1 transcript was first detected in shoot apical meristem and rachi;with the development of the flower,PheDof-1 was expressed in glumes and bracts.The difference is that PheMADS-14 transcript was expressed in floral-organ in moso bamboo,the signal appeared in the parts of cell cleavage,including carpel primordial and stamen primordial,and pollen tube.According to the results of transcriptome and differentially expression profiles for the moso bamboo,a large number of genes information were abtained,and the regulation mechanism of PheDof-1,PheMADS-14 gene were investigated in different developmental stages.This is a solid foundation for revealing the molecular mechanism of moso bamboo flower,and it's a milestone to discover useful genes in flowering of Phyllostachys edulis.
