Mesenchymal stem cells (MSCs) are multipotent stem cells with highly proliferation, self-renewal and multilineage differentiation potential. That can differentiate into a variety of cell types, including:osteoblasts (bone cells), chondrocytes (cartilage cells) and adipocytes (fat cells). MSCs are collected and amplified easily, and their potential of multilineage differentiation was preserved during the long-term culture in vitro. MSCs have special biological characteristics, such as secreting hematopoietic growth factors, reconstructing hematopoietic microenvironment, low immunogenicity, transfecting and expressing and by exogenous gene easily. Many reports indicated that, MSCs were applied in therapy for variety of tissues and organs injury has made significant progress. In this study, the biological characteristics and new research advance on MSCs in treatment of acute intestinal radiation sickness are summarized and discussed.MSCs were isolated from human umbilical cord (UC-MSCs), and were cultured and expansed in vitro. UC-MSCs were characterized by flow cytometric analysis. Cells were positive for CD73, CD90and CD105;but negtive for CD45,CD34,CD11b,CD19and HLA-DR.By using differentiation and culture in vitro, UC-MSCs were differentiated toward osteogenic and adipogenic. After detected by RT-PCT, UC-MSCs can secrete many cytokines, such as SCF, TPO, IL-3, IL-6, FL, M-CSF, G-CSF, GM-CSF, HGF, LIF, VEGF and SDF1.We also study on the therapeutic role of transplantation of UC-MSCs in the mice models of intestinal radiation disease. Human UC-MSCs which labeled with CM-Dil florescence dye, were transplated to mice models of intestinal radiation damage. After that, pathological changes and location of donor cells in intestinal mucosa were observeded by histochemical stain and LSCM (laser scanning confocal microscope). The result show that,there are significant improvement in the general condition and patholog on the mice model after UC-MSCs transplantation. And the UC-MSCs with florescence were found distribute in intestinal lymphoid tissues and epithelial cells, this means MSCs might involved in the repair process of intestinal mocosa.In this thesis,several lines of evidence suggesting UC-MSCs could improve intestinal lesions of mice models significantly. It is also shown that, MSCs hold great promise as therapeutic tools to treat intestinal radiation disease.
