Interventional therapy, which is the main methord to treat coronaryheart disease now, faces restenosis after stenting. There are still manydrawbacks of medical stents, and they indicate that either merelypromoting endothelialization or only suppressing hyperplasia isinsufficient to alleviate restenosis effectively.Here we developed a kind of polyelectrolyte multilayers consistingof PrS and pDNA of tow functional genes using layer-by-layer technique,and it incorporated HGF gene and TGF-β1-shRNA gene into the film.This multilayers can transfect cells, enhancing HGF secretion whilesuppressing TGF-β1secretion at the same time, thus it is able to achievethe goal of promoting proliferation of endothelium cells and suppressingECM secretion synchronously, and may be a new way to treat restenosis.Experiments showed that the polyelectrolyte multilayer could be built upsuccessfully and exhibited good stability under physiological conditions。It was able to transfect3T3cells and made cells express the desired genes.This coating is considered to have the gene therapy function of regulatingthe two growth factors and FN secretion. The experiment of the animalmodel of rabbit had confirmed that the gene-coated stent can prevent of restenosis in vivo.To sum up, this study achieved the goal that two genes were loadedto the polyelectrolyte multilayer films at the same time by the technolodyof layers self-assembly, and it obtained the following results: multilayerfilm could assemble and maintain stability under physiological conditions,and could transfer3T3cells to get the gene therapy effectly whichachieved the level of protein secretion and extracellular matrix secretion.And the experiment of the animal model of rabbit had confirmed that thegene-coated stent can prevent restenosis in vivo.The results of experiments provided evidences in prevention andtreatment of ISR in clinical application. Prevention of restenosis throughendothelial cells planting has shown attractive prospects, with furtherin-depth study that will be opened up to a new area.
