In bone remodeling, the Eph family was involved to regulate the process ofosteoclasts and osteoblasts coordination in order to maintain bone homeostasis. In thisexperiment, effects of lipopolysaccharide from Porphyromonas gingivalis on theosteoblast-osteoclast bidirectional EphB4-ephrinB2signaling were investegated.Therefore, we intend to establish osteoblasts-osteoclasts co-culture system in vitro toobserve the lipopolysaccharide-mediated ephrin-Eph bidirectional signal transductionin genes expression level by real-time PCR and protein expression level by Westernblot and Immunofluorescence technique. Our objective is to clarity the mechanisms ofEphB4-ephrinB2mediated local alveolar bone homeostasis imbalance, and to providenew ideas of prevention from periodontic disease.Conclusion1. RAW264.7cells induced by1α,25(-OH)2VitD3and Dexamethasone can expressephrinB2ligands protein.2.75ng/ml LPS can upregulate the expression of EphB4receptors inosteoblasts-osteoclasts co-culture system.3.75ng/ml LPS has no significant influence or little inhibition on the expression ofephrinB2ligands in osteoblasts-osteoclasts co-culture system.Confirmed Conclution:1. Osteoblasts can successfully be extracted from neonatal mouse mandible byadherent explants method combined with enzymatic digestion method.2. Osteoclasts can successfully induced from RAW264.7cells by1α,25-(OH)2VitD3and Dexamethasone.
