Objective:To investigate the effects of a novel histone deacetylase inhibitor, NKL30on growth of grafted human cervical carcinoma in SCID mice and the expression of Nur77in grafted carcinoma cell.Methods:Cervical cancer models of SCID mice were established by inoculating Caski cell suspension(1*107/ml) subcutaneously into peritoneal surface. These mice were randomly divided into4groups of5each group, injection the different concentrations of NKL30through intraperitoneal treatment. After the treatment32days the mice were killed to remove specimens. We observed the growth of the transplants in SCID mice,measuring the weight and the volume of the transplants. Then we detect Nur77protein by immunohistochemical method.Results:The cervical cancer model in SCID mouse was successfully established:tumor formation was100%. The growth curve of treated mice tumor compared with the control group was significantly inhibited. The tumor weight and volume decreased significantly compared to the control group (p<0.05). Nur77expression showed strong positive expression in50mg/kgNKL30concentrations group. Nur77expression showed positive expression in20mg/kgNKL30concentrations group. And in10mg/kgNKL30concentrations group, Nur77expression showed a weak positive expression. In control group it was negative.Conclusion:1. NKL30inhibits the growth of grafted human cervical carcinoma in SCID mice. 2. NKL30affect the expression of Nur77in grafted carcinoma cell.
