Objective:1. To investigate the effects of glucagon-like peptide-1(GLP-1) on the proliferation of3T3-L1fibroblasts;2. To investigate the effects of GLP-1on the differentiation of3T3-L1fibroblasts;3. To investigate the effects of GLP-1on PPARy and GLUT4mRNA expression during the differentiation of3T3-L1fibroblasts.This study was aimed at characterizing the response to GLP-1of preadipocytes, in terms of proliferation and differentiation, to clarify the function of GLP-1on lipid metabolism and then discuss its therapeutic use from this point.Methods:1.3T3-L1fibroblasts were cultured in the presence of GLP-1of different concentrations (10-11M、10-10M、10-9M、10-8M、10-7M). XTT assay was applied to evaluate the cell growth.2.3T3-L1fibroblasts were induced to differentiate in both treated groups containing GLP-1of different concentrations (10-11M、10-10M、10-9M、10-8M) and the control groups. Oil red0staining method was used to assess the lipid accumulation.3. RT-PCR was applied to measure the GLUT4and PPARγ mRNA levels during the course of differentiation.Results:1. GLP-1attenuated the proliferative activity of3T3-L1fibroblasts especially in higher dose.2. GLP-1inhibited the differentiation of3T3-L1fibroblasts.3. GLP-1exhibited no obvious effect on PPARγand GLUT4mRNA expression during the course of differentiation.Conclusion:Our study discovered that GLP-1inhibited the proliferation and differentiation of3T3-L1fibroblasts for the first time and explained its mechanism of weight loss from the view of adipose tissue.
