Background:The theory of cancer stem cells (CSCs) states that a subset of cancer cells within a tumor has the ability to self-renew, differentiate and proliferate extensively. Besides the traditional methods to enrich CSCs by SP analysis, surface phenotype and serum-free culture, it has been proved that breast cancer cells with these properties of CSC could be enriched by sequential in vivo passage in chemotreated immunodeficiency mice.Objectives:To investigate the effect of chemotherapy on the proportion of CSC-like cells in ovarian cancer cell line OVCAR-3and H8910in vitro. And to investigate the expression of stemness and drug resistance genes, and the drug resistance in SP and NSP cells.Methods:5human ovarian cancer cell lines were test used the Hoechst33342methods, and two of them had SP, detected the drug resistance, the proliferation in vitro and in vivo, and the expression of drug resistance genes and stemness genes. Establish the model to enrich the CSC-like cells by chemotherapy in vitro.Results:1.Some of human ovarian cancer cell line had SP.2.SP showed better durg resistance than NSP.3.The expression of muti-drug resistance genes ware different in SP and NSP.4. The cell cycle of SP and NSP was different.5. The expression of stemness gene Klf4, Oct-4were higher in SP.6.Chemotherapy in vitro can enrich SP cells in human ovarian cancer cell line.7. The enriched SP cell can differentiated to NSP.Conclusions:The Side population of ovarian cancer cell lines partly shows the stemness characteristics. CSC-like cells can be enriched by chemotherapy in vitro.
