Objective: To investigate the function and mechanism of algR gene in Pseudomonas aeruginosa biofilm seeding dispersal.Methods:Deleting the algR gene of PAO1and PA17by using the homologous recombination.The plasmid pVDtacPIL,pVDZ’2R and pEX1.8-rh1AB was each transfected into PSL317and PA17’.The bacteria were cultured to establish a biofilm model, the1,3,5,1,9and12days old biofilms were observed by Confocal scanning laser microscopy with SYTO9/PI viability staining. Real-time fluorescent quantitative reverse transcription-polymerase chain reaction(RT—pCR)was used to detect the expression level of algR gene in PAO1,PA17,PSL317,PA17’,PAOmucA56and homologous recombinant strains,rhlA/B gene in PAO1and PAOmucA56at different time.Observe the swimming motility, swarming motility and twitching motility of PAO1,PA17,PSL317,PA17’,PAOmucA56and homologous recombinant strains.Result:PAO1exhibited seeding dispersal at12th day, PA17at7th day and PAOmucA56at7th day. The expression level of algR gene was highest at11th day in PAOl,at7th day in PA17and PAOmucA56. The expression level of rh1A/B gene was highest at11th day in PAO1. The swimming motility, swarming motility and twitching motility of PAO1was stronger than PA17.Conclusion:The phenomenon of seeding dispersal" was earlier in PA17than in PAO1which was associated with algR gene...
