Objective:Abnormal sex hormones level has been obsearved in obese adolescents these years. Researches of the relationship between obesity and sex hormones are limited by some factros such as ethic, race or experimental condition, but these studies also reflected that the sex hormones level is colsely related to obesity. So the aim of this study was to compare the sex hormones level in boys aged 9 to 12 with different BMI levels, and then used the lipidomics to explore the relationship between obesity and sex hormones level.Content:(1)TC, TG, HDL-c, LDL-c and some sex hormones were detected, and then compare the difference of these indexes in three groups. (2)The Waters UPLC/QTOF/MS instrument combining Progenesis QI software were used to identify the most significant lipid metabolites in different groups and explore the relationship between obesity and sex-steroid hormones.Objects:A totol of 76 boys aged 9 to 12 were included into this study. The age of these subjects is 10.28±0.69 years old. These subjects was divided into normal (NC) group, overweight (OW) group and obese group (OB).Methods:TC, TG, HDL-c, LDL-c, sex hormones were detected by CLIA and ELISA methods.;The analysis procedure were complished by using SPSS 22.0 software. A lipidomics method was establiehed by useing a Waters Acquity UPLCI-Class liquid system combined with Waters Xevo G2-S Q-TOF mass spectrometry system; The indentification and analysis of lipid metabolites were complished by using Progenesis QI software and LipidMaps database.Results:(1)The HDL-c and SHBG levels ware significant lower in OW(p<0.01) and OB(p<0.01) groups. the DHEA levels was significant higner in OW(p<0.01) and OB(p<0.05) groups. The TG level was significant higner in OB group (p<0.01). (2) The Waters UPLC/QTOF/MS instrument combiningProgenesis QI software were used to established a lipidomics method to indentify the most significant different lipid metabolites between NC group and OW&OB group, and 153 lipid metabolites were obsearved by this method including 39 glycerolipid metabolites,92 glycerphospholipid metabolites,23 sphingolopids metabolites.Conclusion:(1)The DHEA levels was decreased and the SHBG was increased in OW and OB groups. (2)A lipidomics method was established to comapre the different serum lipid metabolites between NC group and OW&OB group of 9-12 male adolescents for the first time, and we observed that the glycerolipid metabolites was increased, the glycerophospholipid and sphingolipid metabolites were decreased in OW & OB group. |
