Objective: Blood-brain barrier injury is not only pathology manifestation of cerebralischemia reperfusion injury, but also an important step leading to brain issue injury. Thereserach is to study the protective mechanism of acute cerebral ischemia reperfusion injuryfor rat blood-brain barrier by electroacupuncture.Methods: Rat acute cerebral ischamia model was successfully established by middlecerebral artery occlusion(MCAO). Using electron microscope, observe ultramicrostructureof blood-brain barrier, By immunohistochemistry ABC dye method, observe the express ofMMP-9, TIMP-3, IGF-1, and neuroethology score by Zea long score method. Study theinfluence of electroacupuncture for rat cerebral ischemia reperfusion injury blood-brainbarrier.Results: In model group, astrocyte foot process is Severely oedema, to submitvacuolization alteration, crushing blood capillary, causing lumina narrow, endothelial cellnucleus deformation, cytoplasm uneven, mitochondria swelling cristae breakage, it is thusevident that tight junction is breaking. But in electroacupuncture group, the oedema ofastrocyte foot process is subsidized, blood capillary lumina is unobstructed, endothelial cellnucleus without swelling and deformation, cytoplasm uniformity, mitochondria swellinglessened, tight junction structure is normal. MMP-2, MMP-9 andTIMP-3 is expressed inhippocamp CA1 region neuron in model group, expression location is in cytoplasm, positiveand negative cell is stray distributed. The expression of MMP-2, MMP-9 andTIMP-3 isdecreased in electroacupuncture group, its expression location is in cytoplasm. In modelgroup, the expression of cortex pyramidal cell neuron IGF-1 displaying there is very littlein pyramidal cell nucleus of striatum homonymy cortical pyramidal cell layer, its expression location is in cytoplasm, hard to distinguished with negative cell, positive andnegative cell is stray distributed. The expression of IGF-1 in striatum neuron demonstratingit is not obviously expressed in granulocyte neuron cytolymph. But it is obviouslyexpressed in pyramidal cell peri-nucleus of striatum homonymy cortical pyramidal celllayer for electroacupuncture group. As for neuroethology score, electroacupuncture groupis lower than model group.Conclusion. It is of protcctive role of electroacupuncture for blood-brain barrierresulting from cerebral ischemia injury, meanwhile, it can effective improve pathologyalteration of ultramicrostructure of blood-brain barrier by acupuncture thereby in earlystage. It also can reduce expression of MMP-9, diminish the destroy for blood-brain barrierbasal membrane, increase expression of IGF-1, inhibit apoptosis, joint action to protectblood-brain barrier, reduce brain edema, protect brain tissue.
