The process of wound repair is a comprehensive interaction of repair cells,cytokine and extracellular matrix. The proliferation and apoptosis is an importantbiology phenomenon in the wound repair, which occurs in the healing of normalwound surface and chronic ulcer, this process is regulated by related gene, protein andcytokine. The cell mechanism research on proliferation and apoptosis during thisprocess and the study with its related gene and cytokine will have an active role topromote the manufacture of western and Chinese medicine, clinical dressing andtreatment apparatus...etc.In the wound process, the formation and shrink of granulation tissue is a key stepin wound healing and tissue reconstruction. The granulation contains rich bloodcapillaries, therefore the newbom capillary change could reflect biological mechanismof wound repair. Past studies showed that wound healing is a strict and orderedprocess of regulation. In the initial stage of tissue repair, proliferation, migration andconversion of various reactive cells satisfies the need of healing. As wound’s closure,restoring cells get decreased with apoptosis. The harmony among each kinds of cells omaintains integrality and stability. The formation of capillary vessels brings animportant effect in the final healing of a wound.It is important to keep coordination between proliferation and apoptosis of eachrestoring cells (including fibroblastic cells, vascular endothelial cells (VEC) andinflammation cells etc.). Regeneration of blood vessels is an extremely complicatedhistological and bio-chemical process, which relates to steps such as proliferation andmigrating of VEC. The local environment (neuron-hormone, and various growthfactors and cytokine) produces different effect on it.Wound repair is a pathological process of granulation instead of necrosis cells,and granulation tissue is mainly composed by capillary vessel, fibroblastic cells andinflammation cells, VEC is the main cells of capillary, its function is as following:1) close conjunction of cells, constituteing natural barrier and ensuring normal bloodcirculation;2)secretion of some active materials, such as: PGI2, NO etc., regulatingvasodilation;3)secretion of some chemotactic factors: IL-2 etc., tending to promotecellular infiltration of inflammation cells;4)secretion of some material of coagulationand dissolution, insuring blood circulation. Therefore, VEC function is a key link ofwound and many diseases. In the early stage, VEC could secret many activesubstances to increase supplement of blood and make for wound healing. In the latestage, VEC gets apoptosis as the capillary gets decrease when granulation changesinto scar tissue. The proliferation in the early stage and apoptosis in the late stage arevery important.Traditional Chinese medicine has an unique effect to wound repair, it canstimulate each process in healing. Chuanxiong Rhizome is applied to promote bloodcirculation and remove blood stasis in clinic. Its main effective component isligustrazine, one kind of effective activity alkaloid, whose chemistry structure is fourmethyl paradiazine. Microscopic capsule is a new technique in drug manufacture.Chitosan is used for the membrane of the capsule, which is compatible with biologicalsubstance and easy degraded. So the microscopic capsule of ligustrazine is studied onVEC by flow cytometry, fluorescence microscope and radio-immunology assay toexplore VEC proliferation, apoptosis, gene expression and inflammation mediator.The results showed: the microscopic capsule of ligustrazine has tendency ofpromoting proliferation of HUVEC during a certain time with significant effect at 24h.The expression of Bc1-2 and ICAM-1 is highest at 24h too. The secretion of IL-6 getsto peak at 24h. All this function is slower than the one of ligustrazine itself. Thisindicates the microscopic capsule has function of controlled release and accumulation.And it is beneficial to the wound healing.
