Objective:This study aims to observe effect of acupuncture and Runmuling on dry eye, and explore the mechanism of anti apoptosis and related genes of muscarinic acetylcholine receptor mediated signal transduction pathway.Methods:36 healthy New Zealand rabbits were randomly divided into 6 groups each with 6 rabbits:the blank group, the model group, the pilocarpine group, the rumuling group,the electro-acupuncture group and runmuling and electro-acupuncture group. Atropine group,the pilocarpine group, the rumuling group,the electro-acupuncture group and runmuling and electro-acupuncture group were given bin-oculus with 1% atropine sulfate eye drops four times a day until the end of the trial.But in the fourth day of the experiment, dry eye model succeed. Atropine group was no additional therapy. The pilocarpine group, the rumuling group,the electro-acupuncture group and runmuling and electro-acupuncture group respectively, with pilocarpine, rumuling, electro-acupuncture and runmuling combined with electro-acupuncture. We observed tear secretion and morphology changes of corneal and lacrimal gland of all six groups, acetylcholine content, immunohistochemistry and PCR real-time detection.Results:Schirmer 1, tear film breakup time and corneal staining of western medicine, acupuncture and Chinese medicine, acupuncture and medicine combination therapy were confirmed to promote lacrimal secretion, extend the tear film breakup time and repair damaged corneal epithelium. The effect of the acupuncture and medicine combination therapy was the best. 18 days of the experiment, six groups of lacrimal gland and corneal tissue were used to make laboratory tests. Traditional Chinese medicine intervention group,the electroacupuncture group,Western medicine intervention group appeared some improvement.Acupuncture and Chinese medicine combination intervention group of lacrimal gland and corneal tissue morphology was closest to blank control group. About ach contents, acupuncture and Chinese medicine combination intervention group was significantly higher than atropine model group in day 18. Apoptosis factor Fas, FasL, Bax of semi quantitative in acupuncture and medicine intervention group was significant difference compared with atropine model group. In day 18, M3 AchR gene expression of acupuncture and Chinese medicine combination intervention group was significantly lower than atropine model group; Comparing with atropine model group, MAPK gene expression of acupuncture and Chinese medicine combination intervention group was significantly decreased.Conclusion:Electro acupuncture and traditional Chinese medicine Runmuling can promote lacrimal secretion, keep the stability of the tear film and promote lacrimal secretion, keeping the stability of the tear film and promote lacrimal secretion, keeping the stability of the tear film and the corneal wound heal the corneal by inhibiting the apoptosis of lacrimal gland, promoting the neurotransmitter release and regulating muscarinic receptor mediated signal transduction pathway.
