Research backgroundVaricella-zoster virus(VZV) is commonly regarded as a human herpes virus, Varicella-zoster will appear in the early infection, and the infected viruses can hide in ganglion among the bottom of spinal nerve, viruses re-activation can cause the occurrence of herpes zoster. Segmental blister fluid are caused by viruses from ganglion along peripheral nerve fiber to the corresponding skin epidermis. Regardless of the apparent infection path, virus DNA still can be tested in skin blister fluid,which is in the limits of VZV activation. In order to understand whether there were relationship among VZV DNA load, the pain level of herpes zoster and the patients’ age, We intend to determine blister fluid of patients with varicella zoster virus quantitative DNA.The using of antiviral drugs can effectively control the acute herpes zoster and reduce the occurrence of symptoms and postherpetic neuralgia.Through the study of patients with herpes zoster in the treatment of antiviral drugs famciclovir in one week and two weeks, if there exist pain intensity differences among patients classified into two groups having this treatment on one week, two weeks, four weeks, two months and three months, Maybe it can provide a theoretical basis and a good method for the treatment of herpes zoster patients who are given antiviral drugs for two weeks.The VZV genome contains at least 71 open reading frames (ORFs), and the functions of many of the proteins they encode have been characterized. In comparison of moderate pain with mild pain of patients with herpes zoster, the latter is higher in VZV DNA load. In comparison of severe pain with mild pain, the former is higher in VZV DNA load.2. The result of the DNA load have the statistical significance between over 60 years old and less than 60 years old.Conclusion:VZV DNA load are different among 3 kinds of pain levels. The higher the pain level,the more the viral load. The viral load also exists different among different age groups, age over 60-year-old patients have more viral load than age less than 60-year-old patients.The Part 2 Clinical research of pain intensity comparison among patients with herpes zoster taking famcilovir for one week or two weeks.Objective:Pain intensity scores and incidence rate of postherpetic neuralgia comparison among patients with herpes zoster taking antiviral drugs famcilovir for one week and two weeks.Methods:The pain scores of two groups of patients at different time points were observed by randomized, controlled clinical trial.Results:In comparison the pain intensity during the antiviral drugs famcilovir treatment for one and two weeks, there is no obvious statistical difference in one week, two weeks,four weeks, two months and three months.Conclusion:Pain intensity of herpes zoster patients who is taking famciclovir for two weeks is not better than that of taking famciclovir for one week, In clinical practice, we give patients with herpes zoster oral antiviral drugs for one week instead of two weeks, The economic cost of medicine can be saved and the side effect of medicine can be reduced.The Part 3 Determination of VZV genotypeObjective:To detect the genotype of VZV genotype in 306 hospital of Beijing region.Methods:Varicella zoster virus ORF22, ORF38, ORF54 gene sequence is detected by sequencing method.Results:The results of gene analysis showed that the genotypes of the clinical isolates of 8 strains of herpes zoster virus in Beijing region were J. There is a case of A-G mutation in the 37990 site of ORF22 fragement.Conclusion:The genotype of the clinical isolates of the herpes zoster virus in 306 hospital of Beijing region was J,There is a case of A-G mutation in the 37990 site of ORF22 fragement.
