Objective:To Observe the protective effects of Wen Xin Danon MIRI rats and explore its action mechanism.Methods:1.Make different Wen Xin Dan compatibility proportion groups,control group, model group, and pretreatment group separately work on MIRI rats in order to detect the serum CK and LDH content of rat, the influence of the area of myocardial infarction;examine Wen Xin Dan compatibility relation, finally select the best compatibility proportion Wen Xin Dan group.2. Make the Wen Xin Dan group,control group, model group and LY294002 group separately work on MIRI rats. Then observe the influence of Wen Xin Dan on MIRI rat myocardial tissue pathologic morpHology under the light microscopy using TUNEL method to detect apoptosis index;then detect myocardial tissue Akt, p-Akt, GSK-3 β, p-GSK-3 β (Ser9) protein expression by Western blot method.Use LY294002,the specific inhibitors of PI3K/Akt signaling pathway,to intervent the 1:2:1 group,then investigate whether Wen Xin Dan play an important role on myocardial protection through PI3K/Akt/GSK-3 β signaling pathways.3. Use Flow cytometry instrument to detect changes in mitochondrial membrane potential; detect the Cyt-c, Caspase-9 and Caspase-3 protein expression in rat myocardial tissue by Western blot method and immunohistochemical method.Make intervention of the 1:2:1 group by LY294002, further to investigate whether Wen Xin Dan can inhibit the mitochondrial apoptosis pathway through PI3K/Akt/GSK-3 β signal pathway.Results:1. Each treatment group can significantly reduce the model of rat serum CK, LDH content and reduce the model of rat myocardial infarction area, however, among all the treatment groups, Wen Xin Dan 1:2:1 group’s effect is remarkable.2. Wen Xin Dan could significantly reduce the damage degree of the myocardial tissue of rats, inhibite rat myocardial cell apoptosis and improve the myocardial tissue p-Akt, p-GSK-33 (ser9) protein expression levels. Specificity of PI3K inhibitors LY294002 inhibits Wen Xin Dan’s role.3. Wen Xin Dan can stabilize the mitochondria membrane potential; Significantly reduce the expression of Cyt C, Caspase-9, Caspase-3 protein.Specificity of PI3K inhibitors LY294002 inhibits Wen Xin Dan’s role.Conclusions:1. Different Wen Xin Dan compatibility proportion group can significantly reduce the myocardial enzyme CK, LDH content and reduce myocardial infarction area. Among all the treatment groups,1:2:1 group is the best group to resist myocardial ischemia-reperfusion injury.2. Wen Xin Dan plays a role in myocardial protection by activating PI3K/Akt/GSK-3β signaling pathways to reduce the degree of myocardial tissue damage, inhibit myocardial cell apoptosis and improve the myocardial tissue of p-Akt, p-GSK-30 (ser9) protein expression level.3. Wen Xin Dan can stabilize mitochondrial membrane potential, inhibit Cyt-C release and control Caspase-9, Caspase-3 protein expression by activating PI3K/Akt/GSK-30 signaling pathways.
