Background:Major depressive disorder (MDD) has now become a global impact of mental illness, moreover anti-depression treatment effectiveness is poor, high recurrence rate, which are not only heaven the personal and family burden, but also decreased overall social development. The pathogenesis and neurobiological facts of MDD are remaining unclear; even though our understanding on antidepressant is largely developed by rodent model of MDD, nevertheless double species, rodent and non-human primate, remain large gap including species evolution and other advanced cognition level.Therefore, it is wise to develop one reliable non-human primate model of MDD.Objective:To validated the reliable non-human primate natural occurring MDD model.Method Firstly, by using of simple random sampling to select 10 healthy adult female subjects to performed social isolation induced depression process, to compare the behavior differences between pre- and post- social isolation, to laid solid depressed behaviors references; Secondly, by selecting the subjects with low social ranks and hysteresis traits as the natural occurring MDD samples, and then rule out according to Shively reported criteria,20 candidate subjects were been performed 8 full days observation; Finally, aim to validate the depressed negative impact on subject, and then take the blood sample from natural occurring MDD subjects and controls to perform metabonomics analysis, to compare their metabonomic differences.Results:Social isolation procedure reduced the frequencies of social interaction and locomotion significantly on subjects, moreover this also fade out their attention on other companies and next generation; Similar with this kind of isolation model, natural occurring MDD model presented similar behavior traits including less interesting with food and outside matters, slump posture; meanwhile similar metabolic materials also discovered in depressed rat and patients.Conclusion:Social isolation model provides a solid basis to present depression behavior stablely; Furthermore, natural occurring MDD model, because of social hierarchy, which strength the completion in social context, if subject were not endure this kind of pressures may natural occurring depression. This kind of model is completely natural occurring, without any human intervention, and it is reliable to perform further in-depth investigation on mechanism of MDD.
