Objective; Non-small Cell Lung Cancer (NSCLC) is one of the major malignant tumor, have high invasive and poor prognosis, in recent years, due to environmental factors such as air pollution caused by the onset of Lung adenocarcinoma showed a trend of rising year by year. The role of interleukin-8in the occurrence and development of lung cancer is arousing more and more people’s concern, but few research reports concern of interleukin-8in lung adenocarcinoma in the literature. Our study through detection the expression of interleukin-8in lung adenocarcinoma, and combination with the clinical data, analysis the expression of interleukin-8and lung adenocarcinoma of tumor size, lymph node metastasis and TNM stages, and explore its clinical significance.Material and Methods:58cases are randomly selected as the research objection from patients confirmed of lung adenocarcinoma by intraoperative frozen section examination, take the fresh tumor tissue as the study group, take normal lung tissue next to the cancer as control group. NO35cases, Nl8cases, N215cases. TNM staging phase â… 26cases, phase â…¡13cases, phase â…¢19cases. All fresh tissue after soaking by4%paraformaldehyde liquid preserve, focus on paraffin embedding, each paraffin specimen slice with4μm thickness, and the experiments as follows:(1) HE staining, the microscopic characteristics were observed. (2) By RT-PCR detection IL-8mRNA to compare the genetic level differences between two groups.(3) By immunohistochemical staining detection IL-8to compare the tissue level differences between two groups.(4) Using immunofluorescence staining methods detection IL-8to compare the tissue level differences between two groups.(5) Through ELISA test detection IL-8to compare the portein level differences between two groups.(6) By flow cytometry instrument in the cell level to detect the abnormal expression of cytokines in the organization.(7) Combination with the clinical data, analysis the relation of interleukin-8and lung adenocarcinoma of tumor size, lymph node metastasis and TNM stages.Results:(1) HE staining confirmed58paraffin specimen conforms to the diagnosis of lung adenocarcinoma, and inflammatory cells infiltration for inflammation factor in tumor tissue.(2) RT-PCR detection IL-8mRNA in the gene level confirm the differences between the study group and control group, and significant differences (P<0.05).(3) Immunohistochemical staining in the tissues level confirm the differences between the study group and control group, and significant differences (P<0.001).(4) Immunofluorescence staining in the tissue level confirm inflammatory cytokines IL-8expression was increased obviously in lung cancer tissues compare to control group, and significant differences (P<0.001).(5) ELISA experiments in the protein level confirmed that the inflammation factor had significant difference (P<0.01) in lung adenocarcinoma and normal lung tissue is IL-8.(6) Flow cytometry instrument detection confirms inflammatory cytokines IL-8come from the inflammatory cells infiltrae in lung adenocarcinoma tissues.(7) Combined with clinical pathological characteristics, according to the results of when mass≧7cm, IL-8expression significantly higher (T3, T4group is higher than the T2and T1), with significant difference (P<0.01);T1and T2compared with no significant difference (P>0.05), compared to T3and T4is no significant difference (P>0.05). IL-8expression of lymph node metastasis positive was obviously higher than that of lymph node metastasis negative and significant difference (P<0.01); And with the increase of level of lung cancer lymph node metastasis (NOâ†'N1â†'N2), IL-8protein expression in lung cancer are also increased, with significant difference (P<0.05). Lung adenocarcinoma tissues of IL-8expression level rise, with the progress of the clinical TNM-staging, phase â…¡, phase â…¢ group is higher than phase â… , with significant difference (P<0.01); phase â…¡ compared with phase â…¢, also has a significant difference (P<0.01). By using Pearson correlation analysis, IL-8and T stage, N stage and clinical stage were positively correlated (P<0.01).Conclusion(1) This study respectively from the organization, cell, protein and DNA molecular level proved the expression of IL-8in lung adenocarcinoma tissue were significantly higher than those of normal tissue adjacent to carcinoma, with significant difference (P<0.01); The study has also observed in tumor cells and tumor interstitial infiltration of inflammatory cells can secrete IL-8, but is given priority to with secretion of inflammatory cells.(2) Combined with clinical and pathological characteristics of patients, confirmed that the expression of IL-8and the degree of the tumor size, lymph node metastasis and clinical TNM stages were positively correlated.(3) The above results suggest inflammatory cell secretion of IL-8May participates in the formation of lung adenocarcinoma and progress. Prompt IL-8may be a related to the condition changes and prognosis of patients with lung adenocarcinoma cell factor, can be used as a lung adenocarcinoma condition monitoring and prognosis judgement of one of the indicators. About the role of IL-8in tumor mechanism and clinical application needs further study.
