Study Of Clinical Observation And Immune Regulation Mechanism On Micro-inflammatory State In CRF Improved By “Heidihuang Pills”

Objectives:Due to the effective improvement in correcting renal function, curing renal anemia,preventing tubulointerstitial fibrosis, staying the process of CRF by heidihuang pills,the current study is purposed to analyze the relationship of impairment of immunological function, inflammatory factors,lipid and hormone metabolism disorders.Through the experience on the clinic and laboratory,this research mainly focus on the clinical effect of heidihuang pills on micro-inflammatory state in CRF,try to find out the improving mechanism,seek the best ratio and provide evidences for clinical applications.Methods:1.Clinical Research 60 cases with CRF in accord with diagnostic criteria were randomly divided into heidihuang pills group(n=30) and niao du qing granules group(n=30) and respectively given heidihuang pills or niao du qing granules.Under the same condition,vein blood was collected to detect the level of BUN,Scr,Ccr,CD3,CD4,CD8,CD4/CD8,TC,TG,HDL,LDL,CRP,TNF-ɑ and IL-6 when the patient was fasting plasma.2.Experimental Research successful modeling rats were randomly divided into seven groups(n=15):normal group, model group,niao du qing granules group,lotensin group,high dosage of heidihuang pills group,middle dosage of heidihuang pills group and low dosage of heidihuang pills group.Each group was respectively intervened by niao du qing granules,lotensin,high dosage of heidihuang pills,middle dosage of heidihuang pills,low dosage of heidihuang pills.When the experiment went to the12 th weeks,blood collection was performed from abdominal artery to detect IL-1β、IL-6、TNF-ɑ、IL-10、CRP、ACTH、Cor、TSH HS、LS、PV、FIB、HCT、ESR、ET、CGRP、ATⅠ、ATⅡ. Taking the rats’ thymus and spleen to calculate index of them.Left partial portions of the kidney tissue were collected to observe pathological morphology.Results:Through clinical study,heidihuang pills can inhibit the synthesis and release of inflammatory cytokines(IL-1、TNF-α,CRP)to improve micro-inflammatory state by reducing the level of BUN,Scr,CD8,TC 、 TG and LDL,increasingCD3,CD4, CD3/CD4 and HDL.1.By experimental study,heidihuang pills can increase the index of thymus and spleen with every dosage,pointing its protective effects for thymus and spleen,which can explain the regulation of immune function.2.Heidihuang pills can regulate production and release of IL-1β、TNF-ɑ、IL-6、IL-10、CRP and so on.Heidihuang pills can slow down inflammatory effect of inflammatory cytokines and block Inflammatory process of CRF by reducing the level of IL-1β,TNF-ɑ and CRP,increasing the level of IL-10 and inhibit the secretion of IL 6.High dosage of heidihuang pills has the best treatment.3.Every dosage of heidihuang pills can regulate hormone level of ACTH,TSH,Cor and so on,at the same time,can further cure immune disorders of CRF by inhibiting the secretion of IL-1β、TNF-ɑ、IL-6 and so on,regulating the disorder function of Hypothalamic-pituitary- adrenal axis,increasing the level of ACTH,TSH and decreasing the level of Cor.4.Every dosage of heidihuang pills can regulate the disorder of blood rheology.Decreasing the level of BUN,Sc,inhibiting the production and release of inflammatory cytokines,heightening the level of LS,PV and HCT,lowering the level of PV,inhibiting the synthesis of FIB,slowing down ESR,and adjusting the disorder of abnormal blood rheology and immune disorder function can be main reasons for it to prevent rats’ renal.5.Every dosage of heidihuang pills can regulate the disorder of hemodynamics.Decreasing the level of ATⅠand ATⅡ,reducing the resistance of renal vascular, lowering the level of ET,heightening the level of CGRP,increasingeffective renal blood flow,inhibiting the abnormal rise of IL-1β,IL-6,TNF-ɑ,CRP and so on,reducing the accumulation of Extracellular matrix and adjusting the disorder of abnormal blood rheology and immune disorder function can be main reasons for it to prevent rats’ renal.6.High dosage of heidihuang pills has the best treatment for micro-inflammatory state in CRF.Conclusions:Iimprovement of micro-inflammatory state in CRF by heidihuang pills can have relationship with immunoregulation.The main reasons can regulate of immunologic derangement,inhibit of the synthesis, secretion and release of inflammatory cytokines,slow or block inflammatory effect,adjust the disorder of hormone metabolism,lower Inflammation.Reducing the viscosity of blood and plasma can reduce the deposit of fibrinogen.Lowering the resistance of renal vascular can clear inflammatory cytokines,increase the effective renal blood flow and decrease the accumulation of extracellular matrix to improve renal function,lighten the renal damage and delay renal fibrosis.At the last,High dosage of heidihuang pills has the best treatment for micro-inflammatory state in CRF.
