Objective:By the transplantation of allogeneic human umbilical cord mesenchymal stem cells and the combined use tanshinone IIA. The study on the therapeutic effect of hepatic cirrhosis rats. At the same time to observe the effects of different ways of transplantation on the treatment effect and safety of allogeneic UCMSCs transplantation in rats.Methods:A rat model of cirrhosis was established. The model of liver cirrhosis rats were randomly divided into 5 groups who built UCMSCs transplantation, Control group, tail vein transplantation group, UCMSCs combined with Tan IIA tail vein transplantation group, UCMSCs of hepatic artery interventional treatment group, intraperitoneal injection of UCMSCs group,15 rats in each group. Rats in the UCMSC group were injected via the tail vein with 0.2 ml Dil-labeled UCMSC suspension. Rats in the UCMSC group were injected via the tail vein with 0.2 mL DiI-labeled UCMSC suspension. Intraperitoneal Tan IIA injections (20 mg/kg) were started on the day of UCMSC transplantation in the UCMSC plus Tan IIA group, and continued for 7 consecutive days thereafter. Recording UCMSCs transplantation of rats in each group after eating, drinking, defecation and activities etc. Rats were sacrificed 1 day,3 days,1 month, and 3 months after transplantation and the numbers of DiI-labeled UCMSCs colonizing the liver were determined. Albumin (ALB) and alanine aminotransferase (ALT) levels were measured in venous blood, and mRNA and protein expression levels of human ALB and cytokeratin (CK)-18 in liver tissues were determined by reverse transcription-polymerase chain reaction and western blotting, respectively.Results:Compared with the model group, injected through tail vein, hepatic artery, intraperitoneal injection and UCMSCs combined with Tan Ⅱ treatment group UCMSCs after transplantation. Liver tissues in rats of each group were within the DiI labeled UCMSCs colonization. The rats serum glutamic pyruvic transaminase lowering and albumin increased significantly (P< 0.05). RT-PCR and Western blotting detection results show, rat liver tissue of human CK-18 and albumin mRNA expression after transplantation of human UCMSCs, and Tan IIA combined with UCMSCs in treatment group was higher than that of the pure and the expression amount of tail vein transplantation group (P< 0.05). But after tail vein injection, hepatic artery, intraperitoneal injection of three different ways of transplantation of cell transplantation had no significant difference compared, changes in the rat serum ALT transferase and ALB (P>0.05). AndRT-PCR and Western blotting detection of cell transplantation in liver tissue of rats express human CK-18 and ALB mRNA expression in three different ways and there were no significant differences (P>0.05). Allogeneic UCMSCs transplantation rats eating, drinking, defecating and activities and there were no abnormal before transplantation, rats were no abnormal death.Conclusions:Human umbilical cord mesenchymal stem cells after transplantation can colonization in cirrhotic rat liver, and has the ability to hepatocyte like cell differentiation, so as to improve the cirrhosis rat liver function, Tan IIA combined with UCMSCs transplantation can enhance the transplantation effect to a certain extent.no significant difference between the tail vein injection, hepatic artery and abdominal injection of three the results of different ways of transplantation Transplantation; allogeneic umbilical cord mesenchymal stem cell transplantation in the treatment of cirrhosis of the liver is a safe and effective. Therefore, the subject of prompting the clinical transplantation of stem cells for the treatment of liver disease, can choose the easy operation and trauma of the peripheral vein of liver stem cell transplantation as the preferred method of treatment, and the combined use of Tan IIA can improve the transplantation effect.
