Objective: Discussion SDF-1monoclonal antibody carrying Ultrasound-targetedmicrobubble with the coronary artery transplantation of pig bone marrow mesenchymalstem cells (MSCs) homing relationships and explore bone marrow mesenchymal stemcell transplantation in the treatment of myocardial infarction optimal timewindow.Methods: The39Chinese mini-pigs were randomly selected3as a controlgroup, according to more than36after myocardial infarction,1d、3d、1w、2w、3w、4w six time points were randomly divided into six groups, each6, each experimentalgroup was divided into two subgroups: SDF-1measurement group (n=3) and stem celltransplantation group (n=3). SDF-1measurement and control groups were injectedSDF-1monoclonal antibody carrying Ultrasound-targeted microbubble (Tgroup) andcommon ultrasound angiography agent (Cgroup), the measured parameters to obtainangiography agent of myocardial perfusion parameters (A, β, A×β), after the death byRT-PCR to obtain tissue SDF-1content, stem cell transplantation group were treatedbetween intracoronary injection of BrdU-labeled bone marrow mesenchymal stem cells(MSCs), are on transplant3d animals were sacrificed, the detection of myocardial infarctvolume MSCs homing. Results: Targeted contrast agent in myocardial perfusionparameters (A, β, A×β), the organization SDF-1αmRNA expression and the number ofstem cells homing began to increase both from the first day, to the first seven days of thepeak and then begin to decline. The expression level of the organization SDF-1αmRNAand the number of stem cells homing are correlated (r=0.707, P<0.01); targeted contrastagent in myocardial perfusion parameters A, β, and A×β and the number of stem cellshoming are correlated (r=0.658,0.778,0.777, P<0.05); β (X) and the number of stemcell homing (Y) set up a regression equationY=36.11+17.601X (R2=0.605, P<0.05);A×β (X) and the number of stem cell homing (Y) set up a regression equation Y=50.023+3.348X (R2=0.604, P<0.05). Conclusion: Targeted contrast agent inmyocardial perfusion parameters values, SDF-1levels of the organization and thenumber of stem cell homing after myocardial infarction reached a peak at1week.Targeted contrast agent in myocardial perfusion parameters values can be dynamicallyevaluate variation the number of stem cells homing and SDF-1content time windows, acorrelation between them.
