| Objective: Neuropeptide Substance P(SP), Calcitonin gene-related peptide(CGRP) exists in rat sciatic nerve injury back together root ganglion. This study explores the rat sciatic nerve injury back in root ganglion neuropeptides SP and CGRP expression and meaning, to further explore the neuropeptides in peripheral nerve injury may play a role to provide experimental basis. Methods: The adult male rats were collected(SD) 40, including preparation of 20 SD rats on the left side of the sciatic nerve injury model group(Group A); 10 SD preparation on the left side of the sciatic nerve of rats from the final match model group(Group B); Collected at the same time SD 10 as normal control group(Group C). ①Postoperative 1w, 2w, 4w: observation Group A waist 5 dorsal root ganglion of substance P(SP), calcitonin gene-related peptide(CGRP) positive cell count and mean optical density(MOD), and comparing with Group C.② Postoperative 4w, 12w: Observation Group B waist 5 dorsal root ganglion of SP, CGRP positive cell count and mean optical density(MOD), and comparing with Group C.③ Postoperative 12w: To observe the histopathologic study sciatic nerve in Group B; To observe the anastomosis after sciatic nerve electrophysiological changes; Observation Group B SP and CGRP nerve fibers; Protein gene product 9. 5(protein gene product 9. 5, PGP9.5) nerve fibers. Results: 20 cases of rats on the left side of the sciatic nerve injury model group(Group A), 10 cases on the left side of the sciatic nerve of rats from the final match model group(Group B) and 10 cases of normal control surgical group(Group C) into the result analysis. ①Postoperative 1w: Group A in the dorsal root ganglion of SP and CGRP positive cell count and the MOD is higher than Group C, both have significant difference(p﹤0.05).② Postoperative 2w: Group A in the dorsal root ganglion of SP and CGRP positive cell count and the MOD is higher than Group C both have significant difference(p﹤0.05). ③ Postoperative 4w: Group A in the dorsal root ganglion SP and CGRP positive cell count and MOD Group C with no significant difference(p > 0.05). ④ Postoperative 1w Group A in the dorsal root ganglion of SP and CGRP positive cell count and the MOD is higher than the 2w, both have significant difference(p﹤0.05). ⑤ A set of SP and CGRP in dorsal root ganglia of the mean optical density peaked in postoperative 1w, 4w back to normal levels. ⑥ Group B at postoperative 4w in the dorsal root ganglion of SP and CGRP positive cell count and the MOD is lower than in Group C, there is significant difference(p﹤0.05). ⑦ All landowners Group B at postoperative 12 w in the dorsal root ganglion of SP and CGRP positive expression and Group C has no obvious difference(p > 0.05). ⑧ Group B at postoperative 12 w protein gene product SP, CGRP and 9. 5(protein gene product 9. 5, PGP9.5) showed immune positive nerve fibers, SP and CGRP nerve fibers can be observed. Group B 12 w display sciatic nerve function index has a very good recovery, existing in regeneration of nerve SP and CGRP nerve fibers. Conclusion: SP and CGRP in the varying degree existence with rat sciatic nerve injury back root ganglion. After sciatic nerve injury in rats, SP and CGRP positive expression in dorsal root ganglia in a significant increase in the early, can be used as an indicator of sciatic nerve injury early. SP and CGRP not only involved in nerve injury signal transduction, and in rat sciatic nerve injury in the regenerative repair may play a regulatory role. |
