| AimTO explore the mechanism of anti-AD by Zhisanzhen, the abilities of study, the expression of protein of PP1,PP2 in Hippocampus, activities of AChAT(Choline acetyl enzyme), AChE (Acetylcholinesterase) and GSH-Px in serum of AD-model rats are observed.METHODSThe 80 SD male rats were randomly divided into A, B, C, D four groups in which D group is the control group,20 rats in each group, which were numbered Al~20, Bl~20, Cl~20, D1~20 respectively. D-galactose intraperitoneal injection (50mg/Kg/d) were operated for 6 weeks and A, B and C group were prepared to be the models of AD. The rats in the model group were drilled with dental drill on the skull and injected with Aβ1-40 solution 2ul (10ug).AD rats were divided into three groups, namely the Zhisanzheng group, non acupoint group and model group.1 week after modeling, the zhisanzheng group received acupuncture Shenting and Benshen and non acupoint group received acupuncture at fixed point (non acpoint) under the double ribs of rats. Acupuncture methods: acupuncture with uniform twirling at 180° with 60 times every minute and doing that continuously for 2 minutes by resting 1 minute, all mostl5min. Course of treatments:1 time per day, rest in weekends.20 times totally. The rats in normal group and the model group were given the same treatments, except acupuncture. The brain hippocampus was removed and immunohistochemistry was done form the frontal lobe of the 2mm-8mm coronal slice. To exam the abilities of studies and memory of the rats, Y maze system. was used. The expression of PP1, PP2A protein, AChAT (choline), AChE (acetylcholinesterase) and GSH-Px activities in serum of rats were examed after the experiment.RESULITS1.TRT of each groups at same time:1st,2ed,3rd,4th and 31st day, model group and Zhisanzheng group are greater than normal group (P< 0.05); the 31 day and zhisanzheng group is less than that in the model group significantly(P < 0.05). In the same group, TRT at different time points:(1)Model group on day lcompared with 2,3,4,31 days is significantly different (P< 0.05). 2,3,4,31 days, pairwise compared is almost same(P> 0.05). (2)The normal group at day 1 and day 2 compared is almost same (P> 0.05); and 3,4,31 days compared is significant different (P<= 0.05); day 2 and 3,4 days, the difference was not statistically significant (P> 0.05); and the 31 days compared with statistical significance (P<= 0.05); third,4,31 days, pairwise compared statistically significant (P> 0.05). (3) The zhisanzheng group on day 1 and 2,3,4,31 days compared with statistical significance (P<= 0.05); day 2 and 3,4 days compared statistically significant (P> 0.05); and the 31 days compared with statistical significance (P<= 0.05).2. Immunohistochemistry study showed that compared with normal rats, the expressions of PP1 and PP2A in hippocampus of rats in model group decreased significantly but after the Zhisanzheng intervented, expression of PP1 and PP2A increased significantly, but there is still a significant difference compared with control group3. Compared with the rats in normal group, CHATand AChE activities in hippocampal of the model group rats were decreased significantly. After the Zhisanzheng intervented, the activity of ChATã€AchE increased significantly, but compared with the rats in the control group, there is a significant difference, and compared with the rats in the non acupoint group activities of ChATã€AchE were different significantly.4. Model group and non acupoint group and Zhisanzheng rats’ activity of GSH PX in serum was decreased significantly(P< 0.001) by the statistical analysis. Compared the Zhisanzheng group with model group, GSH PX’s activity in the peripheral serum wasdifferent significantly (P< 0.01) by statistical analysis. Compared non acupoint group with Zhisanzheng group GSH PX’s activity in peripheral serum was different significantly (P< 0.01) by statistical analysis.CONLUSION1.The treatment of Zhisanzhen to AD-model rats can decrease the total reaction time (TRT) and the decrease is different significantly compared with the model group and non acupoint group, which indicates Zhisanzheng can improve the abilities of studies and memory of AD rats.2. The treatment of Zhisanzhen to AD rats may increase the expression of PP1 and PP2A in hippocampus of rats probably.3. The effect of Zhisanzhen may come from the increase of the activities of ChATã€AchE in rat’s brain. Acupuncture can increase the content of acetylcholine and the activities of.AChAT and AChE to make the central cholinergic nervous system in an active state. Through the Zhisanzheng treatment, the speed of acetylcholine synthesis and release are accelerated, the rate of utilization improved significantly and ACh content in brain tissue is improved.4. Zhisanzheng can protect the nervous system from oxidative damage. |
