Objective:To observe the change of the protein expression of ERS and it’s mediated apoptosis in the formation and development of NAFLD and explore the therapeutic effects of exercise and diet modification with alone or combined on NAFLD.Methods:90 Spraque-Dawley rats were randomly divided into nine groups:the 8-week and 16-week control group (C1,C2),the 8-week and 16-week exercise prevention group (E1,E2),the 8-week and 16-week model group (M1,M2), exercise group (EM),dietary modification group (FM),exercise combined with dietary modification group (EFM).Group C1, C2 was fed with normal diet, the other groups was fed with high-fat diet. After 8 weeks, FM Group and EFM group were changed to normal diet. E2, M2 and EM group were continued to feed with high-fat diet. EM, FM and EFM group were respectively intervened by swimming exercise, normal diet, swimming exercise connected with normal diet. We observed the pathological changes in liver tissue and the changes of serum total cholesterol (TC), ariglyceride (TG), alanine aminotransferase (ALT), aspartate aminotransferase (AST) and the protein expression of glucose-regulated protein 78(GRP78),protein kinase R-like endoplasmic reticulum kinase(PERK), eukaryotic initiation factor 2 a(eIF-2a), inositol-requiring Enzyme 1(IRE-1), X-box-binding Protein 1(XBP-1), CCAAT enhancer-binding protein homologous protein(CHOP), the c-Jun n-terminal kinase(JNK) and caspase-12 gene and the positive expression rate of CHOP, JNK, Caspase-12 and the hepatocyte apoptosis.Results:(1) Diffused large bubble-like steatosis and lipid droplets were filled with liver cells in M1 group. The incidence of NAFLD was eighty percent. Compared to C1 group, the indicator levels increased; In M2 group, the incidence of NAFLD was one hundred percent. Compared to C2 group, the indicator levels increased. Compared to M1 group, hepatocyte steatosis was more significant in M2 group. (2) In E1 group, the incidence of NAFLD was forty-four percent.Compared to M1 group, the indicator levels decreased. In E2 group, the incidence of NAFLD was sixty-seven percent. Compared to M1 group, the indicator levels increased, but the range was significantly lower than M2. (3) Compared to M2 group, hepatocyte steatosis was less significant in EM group, FM group, EFM group and the indicator levels decreased.Conclusions:8-week high fat diet induced NAFLD. NAFLD was aggravated with the feeding time prolonged. Endoplasmic reticulum stress was involved in the apoptosis pathway of NAFLD. Exercise had a good effect for the prevention of NAFLD. Exercise and diet modification have a positive effect for the therapy of NAFLD. Exercise combined with diet modification was more effective than simple effect. |
