Cervical cancer is the most common gynecologic malignancies. Global estimates that every year there are about500000new cases of cervical cancer,85%of new cases in developing countries. Cervical carcinoma in situ high age of30to35years, invasive of45to55years, in recent years its incidence has a tendency to young. The clinical and molecular biology performance, both with cancer invasion and metastasis of features, but also to patients bring endless trouble and pain, a serious threat to women’s health and safety. Since the pathogenesis of cervical cancer is not clear, in addition to human papilloma virus infection and cervical cancer is closely related to, the occurrence of cervical cancer oncogene is related, such as c-myc gene overexpression, distant metastasis danger estimates increased six-fold, ras gene expression increased with lymph node metastasis, and the pathogenesis of cervical cancer incidence and spread of tumors and has a very close relationship. Cervical cancer often has local spread and lymph node metastasis, tumor spread to the vagina section, uterine, bladder and rectum are the most common site. Therefore, it is necessary to seek new theoretical guidance and discover new interventions clues.Micro RNA (microRNA, miRNA) are a class of eukaryotic cells about22nt endogenous small non-coding single-stranded RNA, through post-transcriptional regulation of, involved in the regulation of almost all life activities and disease processes. Over the past decade, found that the presence of miRNAs differentially expressed between normal cervical tissue and cervical cancer in the study of a number of pairs of cervical cancer, and is predicting the occurrence and development of cervical cancer have an important relationship. Recently, scholars have found that the detection of miRNAs in plasma and that could become biological markers of cervical cancer. These findings greatly encourage our confidence in miRNAS study of cervical cancer.In this study, using gene chip technology in cervical cancer screening, cervical cancer metastases and normal cervical tissue differences in miRNAs, and found: miR-183in cervical cancer, cervical cancer metastases and normal cervical tissue occur simultaneously falling tone, in metastatic cervical cancer decreased more significantly.While miR-215in cervercal cancer, cervical cancer metastases and normal cervical tissue occur simultaneously rising tone. Current research related to miR-183and miR-215in cervical cancer pathogenesis has not been reported. After qPCR validation findings consistent with the screening results, suggesting that miR-183and miR-215may play an important role in cervical cancer.In order to verify the role of miR-183in the incidence of cervical cancer in this study, respectively, through overexpression and silencing expression strategies investigate the effect of miR-183in human cancer cell function from both positive and negative aspects:1. Cell proliferation analysis:determined by MTT method to detect cell proliferation and PI-FACS method to detect the cell cycle of CASKI. We found that over or inhibit the expression of miR-183had no effect on cell growth.2. Apoptosis research: The results of Annexin V assay demonstrated that the overexpression of miR-183significantly induced the apoptosis of CASKIs, and that the apoptotic rate was decreased when miR-183was inhibited.3. CASKI invasive capability study: Transwell method results show that miR-183has negative regulatory impact on cell invasiveness.These findings suggest that miR-183maybe involved in the procedure of attachment and invasion of the cervical cancer cells.In conclusion, this study has analysed the expression features of miR-183in cervical cancer cell and its faction in CASKI. These findings suggest that down-regulated miR-183expression and up-regulated miR-215expression maybe involved in the development and progression of cervical cancer cell, and may afford some research basis in the target diagnosis and treatment in cervical cancer.
