Aim: To investigate the association between the expression of Oct-4and biological behavior or prognosis in cervical cancer.Materials and Methods: Serum-free suspension culture technologywas used to select suspension of microspheres which can stable clone.Tumorigenicity of suspension of microspheres was identificated inNod/SCID mouse. Microarray was used to detect specific expression ofgenes in suspension of microspheres. The expression of Oct-4wasdetected by immunohistochemistry, and the correlation between theexpression of Oct-4and clinical pathological prognostic indicators wasanalyze in cervical cancer.Results:1.At the similar concentrations the ability of serum-freesuspension culture cervical cancer stem cells to make a neoplasmincreased significantly than cervical cancer cells.The difference of geneexpression profile between suspension and non-suspension ofmicrospheres: the following genes was significantly different between theexperimental and control groups: Stem Cell Differentiation (CD44,Oct-4), Markers Cell Cycle Regulators (APC), Cell Cycle Regulators(MYC) and self-Renewal Markers (MYST2, NEUROG2, SOX1).2.The expression of Oct-4is significantly higher in cervical cancertissues than that in adjacent normal tissues(45.87%vs.11.9%,P=0.001).3.The expression of Oct-4is significantly related to differentiation,clinical stage and lymph node metastasis. 4.The5-year survival rate of patients with Oct-4positive expressionwas lower than patients with Oct-4negative expression (36.7%vs.67.7%,P=0.001).5.Cox regression model analyzed clinical stage, lymph nodemetastasis and Oct-4expression and revealed that they made a differencein cervical cancers prognosis.Conclusion:1.At the similar concentrations the ability of serum-freesuspension culture cervical cancer stem cells to make a neoplasmincreased significantly than cervical cancer cells.2.There were many differences between serum-free suspensionculture cervical cancer stem cells and cervical cancer cells in genesspectrum.Our results showed that Oct-4was highly expressed in cervicalcancer stem cells; Oct-4expression was associated with biologicalbehavior.3.The expression of Oct-4is distinct higher in low pathologicalstages, lymph node metastasis and high clinical Stages stem cells.4.The5-year survival rate of patients with Oct-4positive expressionwas lower than patients with Oct-4negative expression (36.7%vs.67.7%,P=0.001).It can be a potential target for cervical cancer treatment.5.Cox regression model revealed that clinical stage, lymph nodemetastasis and the expression of Oct-4are independent prognostic factorsin cervical cancers (P=0.031;0.012;0.001).
