Objective To screen difference microRNA expression profiles related to HH/GLI1signaling pathway in glioma, and further explore the regulation mechanism of microRNA to HH/GLI1signaling pathway.Methods In this study, we use microRNA gene chip to screen difference microRNA expression profiles related to HH/GLI1signaling pathway in glioma specimens. RT-PCR was performed to verify the reliability of the gene chip. Morever, microRNA expression was confirmed in expanding samples. Bioinformatics method was used to predict microRNA target genes, combined with a luciferase reporter gene vector system to confirm the combination of microRNA and target gene.Results22difference expression microRNA were found after screening, among them9microRNA were upregulated (eg. miR-144), and13microRNA were downregulated (eg.miR-125b). RT-PCR results were in good accordance to the gene chip results. The expression of miR-125b was confirmed by RT-PCR in glioma specimen. The predicted target gene of miR-125b was SMO, which was the key gne in HH/GLI signaling pathway, through bioinformatics methods. The combination of SMO gene and miR-125b was further confirmed by luciferase reporter gene vector system.Conclusions The microRNA expression profile was different in brain gliomas with different HH/GLI1activity. miR-125b was found decreased elavted HH/GLI1activity gliomas, and tageted at the SMO3’UTR.
