Objective To observe the effect of KLN capsules (KLN) and its drug serum on meanfluorescence intensity of intracellular calcium in rat ventricular muscle cells.Methods SD rats were divided into four groups, control group and KLN high-dosegroup, KLN medium-dose group and KLN low-dose group. After intragastricadministration, collect the blood to prepare KLN drug serum. Separate a single ratventricular muscle cell. Using Fluo-3/AM probe to make the cells fluorescently labeledand observe [Ca2+]iwith a laser scanning confocal microscope. Add KLN or its drug serumto extracellular solution between the2nd and3rd scan. Observe the effect of KLN onresting cells’[Ca2+]i and release of intracellular calcium store, as well as the effect of KLNdrug serum on the resting cells’[Ca2+]i.Results①Compared with the control group, KLN high, medium and low dosecould significantly increase [Ca2+]iin resting cell cytoplasm (P<0.05), showing a graduallyincreased trend over time and dose dependence.②In calcium-free Tyrode’s solution,compared with the control group, KLN high, medium dose significantly increase [Ca2+]iincytoplasm (P<0.01), promoting intracellular calcium to release.[Ca2+]iin cytoplasmshowed a gradual increase trend over time. KLN high doses took effect more quickly andsignificantly than medium-dose. KLN low-doses showed no significant differencecompared with control group (P>0.05).③Compared with the normal serum controlgroup, KLN high-dose drug serum and KLN medium-dose drug serum significantlyincrease [Ca2+]iin resting cell cytoplasm (P<0.01), while the low-doses showed nosignificant statistical difference (P>0.05). Conclusion KLN promoted myocardial cell spontaneous extracellular calcium influx,and contribute to myocardial intracellular calcium store release. The former indicates thatthe mechanisms of the KLN anti-arrhythmic effects may be related to the calcium channel,while the latter indicates that the drug may also have a positive inotropic effect. KLN drugserum showed the same effect with KLN.
