PartⅠObjective To establish a model of EV71virus infects human blood mononuclear cells. The pathogenesis of EV71infection was explained at a molecular level in the deep going way. And to investigate the role of the EV71standard virus in the induction of chemokine IP-10in human peripheral blood mononuclear cells.Methods A model of cultured human peripheral blood mononuclear cells was established, cells infected with EV71virus,48h after EV71virus infection, cytopathic effect (CPE) was observed, under inverted-microscope. Using double antibody sandwich enzyme-linked immunosorbent assay (ELISA) detected the level of IP-10.Results After EV71infection, the ultrastructural damage was serious. EV71was amplificated significantly by RT-PCR. Under normal condition, no detected IP-10was founded in PBMC. A high level of IP-10(112.89±12.10;80.06±6.23,respectively) was detected in PBMC48hrs after being treated with the100TCID50EV71standard virus and IFN-y(500U/ml).Conclusion EV71virus could infect peripheral blood mononuclear cells, and caused significant morphological changes. EV71virus could induce a high level of IP-10in PBMC, Which may initiate the process of the immune damage. Part IIObjective To investigate the effect of EV71virus on human peripheral blood mononuclear cells and the possible signal transcription pathway, to explore the mechanism of EV71virus infects human peripheral blood mononuclear cells.Methods The expressions of IP-10and IRF-1in human peripheral blood mononuclear cells stimulated by EV71virus were detected by RT-PCR in both gene and protein level. The role of STAT1activation in the induction of IP-10was evaluated.Results RT-PCR had showed that the expression of IP-10and IRF1of PBMC was detected in both gene and protein level; WESTERN-BLOT indicated that STATl phosphorylation was intiated in the PBMC when stimulated by EV71virus.Conclusion EV71virus could induce a high level of IP-10in PBMC, which may play an important role in the acute immune damage after EV71infection. EV71virus induces IP-10may be activated via JAK-STAT signal pathway. STAT1plays a vital role in EV71infection. Through activation of STAT1, EV71starts the IRF1transcription and then starts IP-10transcription. EV71-induced JAK-STAT signaling pathway activation is one of the pathogenesis of EV71infection.
