Avian Leukosis(AL) is a general designation of a variety of avian neoplastic diseases caused by Avian Leukosis Virus( ALV), which widespread by vertical and horizontal transmission in chicken flocks around the world. In addition to cause tumor,ALV can also cause immunosuppressive. Nowadays, study of subgroub J Avian Leukosis Virus(ALV-J) mainly focus on epidemiological survey, virus identification, virus molecular properties and oncogenic mechanism. However,there was fewer reporst on mechanisms of immunosuppressive about immune cells.In order to investigate whether ALV-J can influence the immune cells differentiation and maturation. We established a method for cultivation and purification of dendritic cells(DCs) from chicken bone marrow in vitro to test the effect in ALV-J infection of DCs. The result revealed ALV-J could infect DCs and replicate on the early stage(day 1-4) of cell differentiation. When the cells further differentiate and change, the ALV-J can not infect DCs. The infection of DCs with ALV-J resulted in lower counts and apoptosis on the late stage of cell differentiation. The m RNA expression of some TLRs and cytokines which are close relative with antigen presentation and immune regulation. The result indicate that the difference observed in DCs resulting from infection with ALV-J may trigger disturbances of antigen presentation and immune response.The integration of ALV-J into the host genome may lead to the deregulation of gene expression. These genes contain functional gene and noncoding gene. The micro RNA(mi RNA) could paly an important regulation on DCs differentiation and maturation. We analyze the mi RNA differential expression in DCs to explore the mechanism aboutapoptosis. The analysis result showed four mi RNA(gga-mir-221ã€211ã€204ã€6651) were correlated with cellular response to endogenous stimulus, antigen processing and presentation, apoptosis and Chronic myeloid leukemia.But the mi RNA expression indicated the antigen processing and presentation was not inhibited. And the reason of apoptosis may be more possible.The GO enrichment and KEGG pathway analysis revealed the other most significant difference functions were with respect to physiological function and metabolism like protein metabolism and glycometabolism and so on. So we preliminary think the ALV-J infection induce the deregulation of mi RNA expression. The most mi RNA which had significant differential expression regulate the substance and energy metabolism.This research showed ALV-J could infect bone marrow mononuclear cells, and affect the cells differentiate into DCs and other types cells by regulating the mi RNA expression aberrantly to disturb the metabolism of cells. This would cause the cells can’t have a normal process of differentiation and maturation. The immunosuppression will occur with lack of mature immune cells. Our findings contribute to the understanding of the molecular mechanism between immunosuppression associated with ALV-J infection. |
