Somatic embryo regenerated system has lots of advantages and it is also a good and attention worth transgenic acceptor system. This research aims to build a regeneration system in vitro of several legume forage species through organogensis and somatic embryogenesis. Meanwhile, the orthogonal design was adopted to comparatively study the difference of plant regeneration system via somatic embryogenesis in several legume forage ppecies materials and MS was used as the elementary medium. Besides, somatic embryos as the genetic transformation receptors in regeneration system were used for the study of genetic transformation in legume forage species. The major results of this study were as follows:1. Genotypes, explants and different exogenous hormone and concentration had obvious effect on embryogenic callus and somatic embryos induction, in which, the genotypes Medicago sativa Martin. cv. Zhongmu No.2, Medicago varia Martin. cv. CaoYuan No.3, Lotus corniculatus L. and Stylosanthes spp. were the optimum genotype for callus induction and somatic embryogenesis.2.There were significant influences of different explants types and different concentrations of the supplements on callus induction in six legume forage species materials, and 2,4-D was the main factor effecting the callus induction, second was explants types. Hypocotyl was the best explant for rapid callus and somatic embryos induction.3. The main factors effecting the somatic embryos induction were NAA and 6-BA, but KT and CH had different performance in different materials.4. In this paper, a comprehensive analysis and a comparison on the difference of plant regeneration system via somatic embryogenesis in several legume forage ppecies materials were given. 2,4-D(2.0-4.0mg/L)+NAA(0-0.1mg/L)+KT(0.5-1.0mg/L)+6-BA(0.1-1.0mg/L) was the optimum callus induction medium and the medium of MS containing NAA(0.01-1.0 mg/L) KT(0-2.0mg/L), 6-BA(1.0-2.0mg/L),CH(1.0-4.0g/L) were the most efficient for inducing somatic embryos.
