Therapeutic Effects And Mechanism Of Traditional Galactopoietic Decoction On Rats With Puerperal Hypogalactia

This study aimed to develop new TCM galactagogue prescriptions for sow, which has a good effect of prevention and treatment on clinical postpartum hypogalactia syndrome. Based on hypogalactia model, screening of the new TCVM galactagogue prescription and dose-effect relationship for the optimal dose were studied by lactation quantity per hour of maternal rats and mass gains in the pups. Therapeutical effect and mechanism of prescription on hypogalactia rats were studied by ELISA method, HE staining method, immunohistochemical staining method, Western Blot method, Western Blot method and fluorescence quantitative PCR method according to serum prolactin changes, tissue structure changes in mammary gland, distribution and tissue level changes of prolactin and its receptors, expression changes of prolactin receptor, beta casein, key protein in synthetic process of lactose and milk fat, cytokines of jak2-stat5signaling pathways and PI3K/Akt/mtor signaling pathways in the mammary gland. Relevant test results are as follows:(1) The optimal bromocriptine dose of hypogalactia model was1.6mg/kg. The preparation, composed of Astragalus membranaceus (25g), Radix Codonopsis (15g), Radix Angelicae Sinensis (10g), Rehmannia Glutinosa (20g), Rhizoma Atractylodis Macrocephalae (10g), Medulla tetrapanacis (5g), Radix Rhapontici (5g), and Glycyrrhiza (5g), had the best effect on treatment of hypogalactia model. The most suitable dose of Cuirutang was1000mg/kg.(2) Compared with the model group, Serum PRL level and tissue PRL level in mammary gland, as well as weight index of mammary gland, significantly increased in Treatment group, which had no significant difference from the normal group. Appearance and organizational structure of mammary gland were obviously restored to a state of lactation.(3) Compared with the model group, PRLR level and gene expression in mammary gland significantly increased in Treatment group, which had no significant difference from the normal group. PRL distribution and PRLR distribution of Interlobular connective tissue in Treatment group had no obvious difference from the model group and the normal group. In mammary gland flocculus, PRL distribution of treatment group was less than that of model group, more than that of normal group. PRLR distribution of treatment group was more than that of model group, and had no significant difference from the normal group.(4) Compared with model group, gene expressions of Beta casein, alpha whey protein, FAS, ACC and SCD significantly increased in mammary gland of treat group, and gene expression of Beta galactosyl transferase decreased significantly in mammary gland of treat group. Compared with normal group, gene expressions of Beta casein in mammary gland of treat group had a significant decrease, and gene expression of SCD in mammary gland of treat group had a significant increase. Gene expression of alpha whey protein, FAS and ACC in mammary gland of treat group had no significant difference from that in mammary gland of normal group.(5) In jak2-stat5signaling pathways, jak2gene expression in mammary gland of treat group was significantly lower than that in mammary gland of model group, and gene expressions of Stat5, cyclindl and CIS in mammary gland of treat group were significantly higher than that in mammary gland of model group. Meanwhile, CIS gene expression in mammary gland of treat group was significantly higher than that in mammary gland of normal group, gene expressions of Stat5, cyclindl and jak2in mammary gland of treat group had no significant difference from that in mammary gland of normal group. In PI3K/Akt/mtor signaling pathways, Aktl gene expression in mammary gland of treat group was significantly higher than that in mammary gland of model group, and significantly higher than that in mammary gland of normal group, gene expressions of PI3K and mtor in mammary gland of treat group had no significant difference from that in mammary gland of both the normal group and the model group, and PTEN gene expression in mammary gland of treat group had no significant difference from that in mammary gland of the model group but was significantly lower than that in mammary gland of normal group.Results above showed that Cuirutang can significantly increase serum prolactin levels, restore mammary gland to normal state from degraded state, increase significantly distribution and protein level of prolactin and its receptors in mammary gland tissue, significantly improve expression of beta casein, key protein in synthetic process of lactose and milk fat. Therapeutic effect of Cuirutang in mammary gland mainly acted through jak2-stat5signaling pathway, had no obvious relation to PI3K/Akt/mTOR signaling pathway, except only Akt pathway.
