Growth hormone (GH), synthesized and secreted by cells in the anterior pituitary, is an important polypeptide hormone which can improve growth and development of organism, muscle development and metabolic regulation. This report mainly introduces the work about the research of transgenic pig at post-doctoral research station in recent years. Firstly, we have modified the GH gene and constructed the GH overexpression vector with different introns. The result revealed that partial introns could improve the expression activity of GH gene. The inducible expression vectors and integrated inducible vector with GH gene containing intronl,2have been constructed and the inducible activity and expression efficiency at cell levels have been detected with PCR, QRT-PCR and western blot methods. The integrated GH controllable expression vector was transfected into PEF cells. After screened with G418, the cell lines were as doner cells to prepare several transgenic pigs with handmade clone method. Ear-tip fibroblasts separated from transgenic piglets and the foreign gene was detected. The result showed that the pTTGH have been prepared successfully.Main conclusions:1. The higher expression actively recombined pig GH gene were Screened out;2. The integrated GH expression controllable vector has been constructed;3.
