With the rapid development of society, people require for higher quality and safety tobacco products, and more emphasis on tobacco additives. In this paper, on the basis of the actual situation of China’s tobacco additive industry, and the methods of safety evaluation for tobacco additives, combined with modern separation and purification technology and toxicological evaluation, the identification and evaluation methods for some endogenous risk substances of natural flavors were established. The chemical constituents of the three perfume plants(lysimachia foenum-graecum hance, Tussilago farfara L. and Ziziphus mauritiana Lam)were studied. And their application to the tobacco industry was evaluated.In the chapter 1, the research progress on the safety evaluation of natural flavor used in tobacco were summarized.In the chapter 2,the identification method of endogenous risk substances of natural spices was established. Methods for quantitative analysis of phenol and benzopyrene that produced by tobacco additive pyrolysis were established.In the chapters 3-5, the method for identification of the risk substances in natural flavor was used for research on the security risk identification and the process improvement of three plant extracts(lysimachia foenum-graecum hance, Tussilago farfara L., and Ziziphus mauritiana Lam). The results showed that the extract of lysimachia foenum-graecum hance and Tussilago farfara L. could not be used for the tobacco additives directly. Thus, the risk parts of the two plant extract that did the harmful effect to the security were tested further, and the specific compounds were separated and identified. By the improvement of processing technic, the risk part of lysimachia foenum-graecum extract have been removed, and the final extract was proved to be safe and can be used as tobacco additive without the loss of odor.The study on the chemical constituents of three plants(lysimachia foenum-graecum hance, Tussilago farfara L., and Ziziphus mauritiana Lam.) was carried out. By variety of chromatographies over various materials,62 compounds including 10 new compounds, were isolated from these three species. Their structures were elucidated, and covered triterpenoid saponin type, sesquiterpene type, flavonoid glycoside type and furoic acid type.This study has enriched the chemical and pharmacological activity research of there three plants (lysimachia foenum-graecum hance, Tussilago farfara L., Zizyphus mauritiana Lam.). It’s also provided scientific basis for the further exploitation and utilization of these three plants. The research also provides accurate and objective data to the safety evaluation and normative use of tobacco natural flavor.
