| Acrylamide (AA) is known to be a neurotoxic, genotoxic, and a potentially carcinogenic compound, which existed in heat processing foods widely. Studies had shown that the addition of Glutathione (GSH) could inhibit the formation of AA dramaticlly. In this paper, the reaction products and reaction pathways of AA formation in Asn/Glc/GSH model systems was studied. The main conclusions were summaried as follows:1. The effect of AA formation by GSH addition were studied in Asn/Glc and Asn/Glc/GSH model systems. Results showed that the addition of GSH could inhibit the formation of AA dramaticlly. And the inhibition effect could be influenced by the dosage of GSH addition and the initial pH condition. According to the result, the inhibition of AA by GSH was attributed to the intervention of AA formation, competitive reaction for Glc and elimination reaction of AA.2. Identify the main reaction products in aqueous Asn/Glc/GSH model system heated at 180℃ for 30 min. Four stable reaction products were identified as C10H18N2O8, C16H27N3OnS, C6H12N2O3S and C13H22N4O7S, respectively, by QTOF. The possible competitive reaction for Glc and elimination reaction of AA were proposed based on the structure of the reaction products.3. The UPLC-MS/MS method for simultaneous determination of AA, GSH, GSSG, y-Glu-Cys, Cys-Gly, Gly, Cys, PCA, DFA, DFC, DFG, DFGSH, AA-Cys, RP-1 and AA-GSH in Maillard model systems was developed. It was convenient to analyze 16 compounds in 4.5 min with good linearity. It could be used for simultaneous determination of compounds in Maillard reactions to trace their change of concentration.4. The concentrations of the compounds related with AA were studied in Asn/Glc (1:1), Asn/Glc/GSH (1:1:1/8), Glc/GSH (1:1/8), and AA/GSH(1:1) model systems. The effect of addition of GSH on the formation of AA was investigated. According to the result, the inhibition of AA by GSH was attributed to the intervention of AA formation and elimination reaction of AA.5. In order to improve the mechanism, the Asn/Glc/GSH (1:1:1) model systems were built. Results showed that inhibition mechanism of AA by GSH was mainly attributed to the competitive reaction between GSH and Asn for the consumption of Glc and the the low pH value. |
