MicroRNAs (miRNAs) are a new class of non-coding RNAs which regulate gene expression at post-transcription level and play roles in development, tumorigenesis, embryonic stem cell differentiation apoptosis etc. Many miRNAs have been found to participate in skeletal muscle development in recent years.miR-483-3p was highly expressed in fetal liver, it was located in the intron of IGF2. Up to now, the funtion of miR-483-3p in myogenesis was not very clear. In this study, the myoblast cell line C2C12was used for functional analysis of miR-483-3p in vitro. We try to explore the role of miR-483-3p in myogenesis and identify the new targets of the miRNA. The results are as follows:The expression level of miR-483-3p in muscle tissue is significantly higher than other tissues, and the expresssion trend of the miRNA is similarly to the mRNA of IGF2gene in leg muscle from birth to adulthhood. There is a binding site of miR-483-3P in3’UTR of SRF gene by luciferase repoter analysis, and SRF is identified as a target gene of the miRNA by western blot.The results of CCK-8and xCELLigence detection indicate the overexpression of miR-483-3P can promote proliferation of C2C12cell which is a mouse myoblast cell line. The expression of myhc-2d and myogenin, which are two marker genes of myoblast differentiation, and the ability to fuse and form myotubes are repressed whith miR-483-3p overexpression.Based on these results, we inferenced that miR-483-3p play an important role in proliferation and differentiation of myoblast and in the regulation of myogenesis and muscle development via directly targeting SRF.
