Ultrasound-modulated optical tomography (UOT), which has the advantages of optical contrast and ultrasonic resolution, is a promising noninvasive tomography in biological tissue. Aiming at the problems existing in UOT, some new views and means from theoretical research, technological path and clinical applications are presented as follows:1. The propagation of ultrasound-modulated scattering light in multi-layer biological tissue was studied with Monte Carlo simulation. The results of Monte Carlo simulation are verified with an equivalent experiment. All the results show that the modulation depth of modulated light is only dependent on the optical and ultrasonic properties of the media in the ultrasonic, focal field,so modulation depth is a better physical parameter for image reconstruction and processing.2. An analytical solution about the scattering light intensity modulated by ultrasound in scattering medium based on diffuse theory is presented. An experiment of the incoherent light modulated by ultrasound was obtained for the first time, which provides a major supplement for research on the mechanism of the ultrasonic modulation of light in scattering media.3. Some problems were discussed comprehensively and detailedly about the ultrasound-modulation optical imaging system, including the effect and influence of aperture in imaging system and how to choose the parameter of ultrasonic probe. And a better scheme is propounded to enhance the signal to noise ratio and sensitivity of the imaging system.4. Fast Fourier Transform (FFT) was used in the digital frequency analysis of modulation signal. Compared with the peak-to-peak voltage and modulation depth of ultrasound-modulated signal, the in-frequency signal is not better than the in-time signal for image reconstruction and processing.5. A method for noninvasive measurement of blood glucose levels was presented using ultrasound-modulated optical technique. The results show that modulation depth increase linearly with the increase of glucose concentration in intralipid solution, and this method has good measurement sensitivity, which substantiates the feasibility of this method in measurement of blood glucose.
