Bone repair remains a clinical challenge. With the development of tissueengineering, a more ideal technique for bone repair is possible. Tissue engineering isan interdisciplinary science of co-development and combination of modern cellbiology, biomaterial science and engineering. In the light of present knowledge, theformation and repair of bone with tissue engineering require studies on source ofosteoblast, scaffold materials and construction of tissue engineering bone in vitro. Thepresent study is based on these requirements and contains: Improvement on culturecondition for osteogenic transformation and osteogenic potential of adult bonemarrow stromal cell (BMSC), surface coating of true bone ceramic (TBC) andconstruction with BMSC.Osteoblast from bone marrow is the optimal seed cell, but for the heterogeneity of(BMSC), the culture condition for osteogenic transformation of BMSC was to beimproved; biocompatibility and growth of cells on artificial carrier, the osteogenicpotential of adult BMSC to be further researched. BMSC was derived from adultpatients, co-cultured by dexamethathone and Danshen. The result showed thatDanshen cloud significantly eliminate the inhibitive effect of dexamethathone onproliferation of BMSC, and could improve the osteogenic transformation dramatically.Cultured in 1640 medium containing dexamethathone (10-8mol/L), Bglycerophosphate(10mmol/L) and Vitamin C (50μg/ml), could form mineralized bonetissue in vitro.TBC was an ideal scaffold in bone tissue engineering, with natural bone trabecula,continuous trabecular spaces, porous structure, and strength of bone, which artificialscaffold could not mimic. It exhibits excellent biocompatibility, osteoconduction andbiodegradability. Surface coating with drug-loaded calcium phosphates cement (CPC),enabled it to possess osteoinduction. BMSC was seeded onto blocks of modifiedTBC, cells and TBC composite were entopic implanted. The result showed that cellswere attached to modified TBC and grew favorably, new bone was found in TBCblock after implantation.
