Abstract/Summary:
Purpose:To evaluate the effects of transplantation of hepatocyte growth factor (hHGF)-transgenic mesenchymal stem cells(MSCs) combined with core decompression for osteonecrosis of femoral head in rabbits.Materials and Methods.â‘ MSCs were harvested from iliac of the rabbits,then isolated, identified and serial subcultivated.Adenovirus recombined hepatocyte growth factor was used to modify MSCs.Secretion of HGF from MSCs was observed.â‘¡Micro-CT and gross measurement were used to evaluate the anatomy of proximal femur of rabbits to see the feasibility of core decompression with Kirschner wire.Histology of was proximal femur also assessed.Indian ink was injected into the femoral head under X ray guidance to evaluate the diffusion mode of cell suspension.â‘¢Ninety rabbits were injected with lipopolysaccharide (LPS) 10ug/kg,which is followed by 3 injections of methylprednisolone 20mg/kg.Survival animals were divided into 4 groups and evaluated with blood fat and body weight.Group A was used as control,while animals of group B,C. and D were injected 2mL of saline,MSCs suspension,or MSCs /Ad-HGF suspension through core decompressing bony tunnel,respectively.Rabbits were sacrificed 4 weeks after injection and evaluated with Micro-CT,histology,and VEGF immunohistochemical methods.Results:â‘ MSCs could be cultured and differentiated in vitro.When MOI=150,Ad-HGF could transfect MSCs with high efficiency.MSCs could secrete HGF for 12 days.â‘¡The fossa trochanterica of rabbit was deep.The smallest diameter of femoral neck was larger than 4mm, which means 2mm Kirschner wire could be used for core decompression.Indian ink was well-distributed.â‘¢Group D showed superiority over the other 3 groups on parameters of Micro-CT and histology.Immunohistochemical study showed active vascular proliferation of Group D.Conclusion:MSCs /Ad-HGF suspension injection after core decompression could improve bony repair and vascular reconstruction in the osteonecrosis area.
