Recent researches show the extrinsic coagulation correlates to many pathological progress , such as the inflammation reaction ;atherosclerosis ;cancer genesis and metastasis, et al. Our previous researches have revealed that TF expresses in the colorectal cancer, and promotes the metastatic and invasive properties of the cancer, in vivo and in vitro. Most of the studies focus on the molecule TF, however, as an important part of the TF/Fâ…¦a complex ,Fâ…¦a plays an essential role. And this research on Fâ…¦a present the ectopic synthesis of this coagulation protease in colorectal carcinoma and the correlation with the metastasis to the liver and lymph nodes. Furthermore, we found hypoxia can up-regulate the expression of Fâ…¦a mRNA in 3 colorectal cancer cell lines.Objective: To verify the ectopic synthesis of active Fâ…¦in colorectal cancer, and investigate the correlation of the Fâ…¦ectopic expression with the cancer liver/lymph nodes metastasis. To detect the contribution that hypoxia induces to the Fâ…¦a expression of the 3 colorectal cancer cell linesMaterials and methods: Take the colorectal cancer tissue and normal mucus from the recent 45 operated cases and 3 colon cancer cell lines as the object. Detect the cancer tissue, normal mucus and cell lines with the RT-PCR respectively to find out the Fâ…¦a mRNA expression and contrst the expressive deference of the cancer tissues in various Dukes stages by Real-Time RT-PCR. The protein synthesis in cancer tissue is verified by the immunohistochemistry and the expression variance compared by Western-Blot. Using the semiquantitative RT-PCR to compare the Fâ…¦mRNA expression of Results: The ectopic expression of Fâ…¦a mRNA in colorectal cancer tissue and cell lines is positive , nevertheless is negative in normal mucus. Real-Time RT-PCR reveal the variance of the Fâ…¦a mRNA expression in colorectal cancer in deferent Dukes stages. The volume increase from stage A to D , especially the cases in the stage D or with lymph nodes metastasis ,the volume is significantly deferent from that of other stages. Moreover, hypoxia can also promote the Fâ…¦a expression in 3 distinct tumor cell lines, comparing with that of the cell lines cultured in common condition.Conclusion :Colorectal cancer tissue and cells can express active Fâ…¦mRNA and synthesize the protenase .The expression volume is relative with the Dukes stages and lymph node metastasis prompting that it may enhance the tumor invasive and metastatic capability, however, it is independent from the number of metastatic lymph nodes; Hypoxia can promote the Fâ…¦a expression in cancer cells.
