Bladder cancer is the most commonly seen carcinoma in urinary tract and a disease threatening human health severely.The morbidity and mortality of bladder carcinoma in our country are always increasing. FGFs and their receptors are a group of molecules who play an important role in modifying cell growth and differentiation with diverse biological activity.The FGF/FGFR signal has been proved to be associated with kinds of tumor development.Illustrating their role in bladder cancer is of great importance to help diagnosing and treating.PartⅠThe express of FGFR2 in bladder transitional cell carcinomaPurpose:To evaluate the effect of FGFR2 to the development of bladder cancer.Method:The expression of FGFR2 in 30 bladder cancer specimens was evaluated by immunohistochemical detection and Western blot.Result:The expressing level in cancer was lower than which in the normal bladder epithelium.(P<0.05)The expressing level was higher in the well differentiated group than the poorly differentiated group,lower in the invasive group than the superficial group and lower in the recurrent ones than the no-recurrent ones.Conclusion:The expression of FGFR2 is negative related with the severity of bladder cancer,it may be a tumor suppressor in the disease. PartⅡThe effect of Sex hormones on the growth of bladder cancer cell line RT4Purpose:To evaluate the effect of sex hormones on the development of bladder cancer.Method:The expression of ERa,ERβand AR in RT4 cell was detected by immunohistochemical way.β-E2 and testosterone of gradient concentration was added to the culture media respectively.After a certain time,the growth of each group was detected by MTT method.Result:No statistical different was observed between anyβ-E2 group and the control after 24 hr's culture,but in the two testosterone group,1×10-7mol/L and 1×10-6mol/L,stimulated growth was obtained.After cultured for 48 hrs all the groups of sex hormones showed more growth over the control but the estrogen groups grew less than the testosterone groups.Conclusion:Both estrogen and testosterone can stimulate the growth of bladder cancer but the testosterone is more effective.PartⅢEffect of sex hormones on the expression of FGFR2 in RT4 cellsPurpose:To test the effect on FGFR2 expression in bladder cancer. Method:β-E2 and testosterone of gradient concentration was added to the culture media of RT4 cells respectively and cultured for a certain time. After that the FGFR2 mRNA level was evaluate by RT-PCR.Result:The FGFR2Ⅲb express was higher in the 2β-E2 group of 1×10-7mol/L and 1×10-6mol/L after cultured for 24hr.After 48hr the 2β-E2 group of 1×10-7mol/L also showed higher expression of FGFR2Ⅲb express.But FGFR2Ⅲc did not change in any group for both culture time. Conclusion:The effect of estrogen on bladder cancer growth may be performed by a way with the express of FGFR2Ⅲb.
