Acupuncture Of Different Acupoints On Gastric Motility Of Rats With Bi-directional Regulation Effect

Objective:Although the effect of acupuncture on the body is miscellaneous,it is a kind of positive effect of bidirectional regulation all in all.It will carry out the function of preventing disease,treating disease and enhancing immunity through triggering the body's inherent adjustment system(nerve,endocrine,immune system). But the bidirectional regulation effect of acupuncture hasn't been researched completely.This research used the methods of recording rats' visceral nerve center neur mono-cell action of nucleus of solitary tract and gastric motion to find the effect of bidirectional regulation of reinforcement and restraint in neur action of nucleus of solitary tract by acupuncture.The influence of the discharges of NTS neurons and gastric pressure responding to the acupuncture at "Zusanli","Neiguan","Zhongwan","Qihai" point were observed when gastric motion reinforced and restrained.On the base,we researched the regulation function about the recovery of organs' function and two kinds of neur action of nucleus of solitary tract by acupuncture in the acupoints of reinforcing gastric motion and restraining gastric motion to probe into the bidirectional regulation function caused by acupuncture in the acupoints and the nerve mechanisms of causing this effect.Methods:Forty-five healthy SD rats of male sex,weighing 300~350g,were anesthetized with 10%urethane by intraperitoneal injection(1.0~1.2g/kg).Drill a hole in the cranium position of NTS(A-11.3~-14.3mm;L0~2.3mm;H 4~7mm) by desk-top tooth drillmobile according to the stereotaxic atlas of the rat(Paxinos and Watson,1998).Adopting electrophysiological methods,the extracellular discharge signals of neuronic action in NTS were recorded with glass microelectrode.The discharge signals of neuronic action were amplified second order by microelectrode amplifier,Vc-10 biophysics amplifier(AVB-10)and displayed on the oscilloscope, and then put into the computer data acquistition system of PowerLab.The gastric motion was recorded by putting a saccule in the rat's stomach.The gastric pressure was put into the pressure capsule from cannula through three-way joint equipment. The hydraulic pressure signals were transformed into electric signals,which were amplified by biological amplifier and put into the computer data acquistition system of PowerLab.Expose the left vena cervicalis and the common carotid artery.The venous cannula was directly connected with the syringe for vein medication.The arterial blood pressure was put into pressure capsule from arterial cannula through three-way joint equipment.The hydraulic pressure signals were transformed into electric signals,which were amplified by carder amplifier and put into the computer data acquistition system of PowerLab to record blood pressure and monitor the rat's status.The influence of the discharges of NTS neurons and gastric pressure responding to the acupuncture at "Zusanli","Neiguan","Zhongwan","Qihai" point were observed in rats' normal status and the condition of intravenous injection Atropine or Metoclopramide.Histology localization was performed after the record of neur mono-cell action of nucleus of solitary tract.The data whose record locations were out of the NTS were cancelled.The useful data were analyzed with the computer data acquistition system of PowerLab and the analysis software of Chart5.0 for Windows, and be processed with SPSS13.0 statistical package.The numerical value of pre-acupuncture,post-acupuncture and difference value in each group were expressed as mean±standard deviation((?)±s)by descriptive statistics.The numerical value of pre-acupuncture and post-acupuncture were analyzed with paired-sample t test to observe the influence of NTS neuron discharges and gastric pressure by acupuncture. The numerical value of pre-medication and post-medication were analyzed with paired-sample t test to observe the acupuncture effect after administration.The statistics standard of significant difference is P<0.05.Results:(1)The influence of the discharges of NTS neurons responding to the acupuncture at "Zusanli","Neiguan","Zhongwan","Qihai" point were observed in rats' normal status and the condition of intravenous injection Atropine or Metoclopramide.The results indicated that acupuncture at "Zusanli","Neiguan","Zhongwan","Qihai" all activated the discharges of NTS neurons remarkably(P<0.05).There were no remarkable differences in the activation effect of the increase of NTS neurons discharge activity caused by the acupoint stimulation in the limbs and the acupoint stimulation in the abdomen.It could obviously activate the discharge activity of NTS neurons by acupuncture at different acupoints in the rat with different status.Although the spontaneous activity of the background discharges increased a little after intravenous injection Atropine or Metoclopramide,there were no statistical differences in the background discharges in comparison with those of the normal rat (P>0.05).(2)The influence of the gastric pressure responding to the acupuncture at "Zusanli","Neiguan","Zhongwan","Qihai" point were observed in rats' normal status and the condition of intravenous injection Atropine or Metoclopramide.The accommodation effects of the rats' gastric pressure were different in type and intensity by acupuncture at "Zusanli","Neiguan","Zhongwan","Qihai" respectively. In the normal status,acupuncture at "Zusanli","Neiguan" could obviously increase the gastric pressure(P<0.01),and the gastric pressure obviously decreased by acupuncture at "Zhongwan","Qihai" point(P<0.01).When the gastric motion restrained after intravenous injection Atropine,the gastric pressure increased obviously by acupuncture at "Zusanli","Neiguan" point(P<0.05),but acupuncture at "Zhongwan","Qihai" point could still remarkably depress the gastric motion(P<0.01).When the gastric motion reinforced after intravenous injection Metoclopramide, acupuncture at "Zusanli","Neiguan" could still promote the gastric motion obviously (P<0.01),but the gastric pressure decreased by acupuncture at "Zhongwan","Qihai" point(P<0.05).Conclusions:The function of adjusting gastric motion by acupuncture at "Zusanli","Neiguan","Zhongwan","Qihai" point all have close relation to the activation of NTS neurons.Acupuncture at "Zusanli","Neiguan" can obviously promote the gastric motion,and acupuncture at "Zhongwan","Qihai" point can obviously restrain the gastric motion.The above regulation effects have relationship with the activation of NTS neurons directly or indirectly or activating the segmented sympathetic nerve of the alimentary system.When the excitability of the vagus nerve predominates,it causes the promotion of gastric motion,and when the excitability of segmented sympathicus predominates,it causes the restraint of gastric motion,which can keep a balance status of autonomic nerve under different situation and educe the optimum of two-way regulating effect.The bidirectional regulation effect of acupoints doesn't surely indicate that the same point has bidirectional regulation function to internal organ in the different pathologic state.Maybe the synthetic action of different points is to adjust the functional activity of body in good regulation and make it in normal status of balance and homeostasis.
