1 Amplification and Elemental Bionomics of Rabbit Adipose-derived Stromal CellsObjective To isolate and amplificate the rabbit adipose-derived stromal cells and study the elemental bionomics of rADSCs.Methods Four apinoid albino rabbits were selected and the adipose tissue in inguinal region was taken out and digested by collagenaseⅡ.The ADSCs were amplificated by paries-sticking methods.The growth kinetics of the ADSCs were measured and the effect of EGF on ADSCs were detected.The stromal cells sufficial antigen CD44and CD45was detected by immunocell fluorometric method.Results The cultured ADSCs go down to posterity from P0 to P8 and the doubling number of each generation showed no descent tendency.The EGF presented marked proliferation promoting effection on the ADSCs.The ADSCs expressed CD44by showing red fluorescence under the fluorescence microscope and CD45expression was nearly negative.Conclusions Isolating and amplificating the ADSCs from adipose tissue were convinent and easy. The rADSCs not only had the elemental bionomics of stem cells but also could be amplificated steadyly to generate enough cells.The ADSCs can be regarded as a hopful seed-cell resource in tissue-regeneration project.2 The Inducing Research of insulin-secreting function on Rabbit Adipose-derived Stromal Cells in VitroObject To study the possiblity of rabbit adipose-derived stromal cells induced and differentiated to insulin secreting cells in vitro.Methods The rADSCs were isloated by wall-sticking method and were induced by GLP-1 and Nicotinamide after being cultured and passage.The DTZ staining experiment and glucose stimulating experiment were performed on the rADSCs after the inducing procedure.Results The rADSCs being induced gathered into clumps and presented red colour by DTZ staining. The insulin secreting under glucose stimulating in vitro was positive.Conclusions The rADSCs have the potentiality to be induced into insulin -secreting cells under certain conditions in vitro.3.Comparation of Inducing Type 1 Diabetes Models on Rabbits by Streptozotocin and AlloxanObjective To compare the effect and character of inducing type 1 diabetes models on rabbits.Methods The STZ and ALX of different concerntrations were injected into the rabbits through auricle edge vein and abdominal cavity to induce the type 1 diabetes models.The blood glucose concerntration and body weight of the rabbits were detected and the glucose tolerance tests were performed on the models. The effection and character of inducing type 1 diabetes models on rabbits by the different methods and drugs were analyzed according the datas.Results The blood glucose concerntration of the rabbits were obviously affected by STZ and ALX(P<0.05).But the inducing effect of STZ was feebler than that of ALX by the same injection methods and the blood glucose concerntration of the rabbits in intravenous injection group was much higher than that of in peritoneal injection.The glucose stimulating responses in treating groups were attenuated in different degrees in glucose tolerance test.Conclusions STZ could be regarded as the reasonable and safe inducing drug by intravenous injection while the ALX should be used as the inducing drug by intravenous injection.4.The Therapic Research on The Type 1 Diabetes Rabbit Models Treated by ADSCs TransplantionObjective To study the the affection on blood glucose concentration(BGC)of rabbits after ADSCs transplantion.Methods The diabetes rabbit models were divided into ADSCs-transplating group through vein,ADSCs-transplating group through arteria pancreaticoduodenalis and control group without ADSCs transplantion.The changes of BGC after the transplantion were caculated.Results The BGC of ADSCs-transplating group through vein descended but the changes were not obvious while the BGC of ADSCs-transplating group through arteria pancreaticoduodenalis decended more obviously.Conclusions The ADSCs-transplanting showed the positive affection on the BGC of diabetes models and the target transplantion through arteria pancreaticoduodenalis could conduce to improve the affection.
