| Objective: To evaluate the effect of Pingganxifeng/Huoxueyishen (PGXF) therapy on the Parkinson Disease (PD) rats, and study the mechanism and biological basis of PGXF method.Methods: We studied the protective effect of the PGXF therapy (a traditional chinese medicine therapy), against 6-hydroxy-dopamine (6-OHDA) induced neurotoxicity ina rat experimental model of PD. L-dopa (LD) served as positive drug. In the experiment, we observed the behavioural change, quantification of Tyrosine Hydroxylase-positive neurons, local anti-oxidation system, dopamine receptor 2 (DR2) mRNA expression and DR2 activity.Results: (1) Compared with themodel rats and those were treated with LD therapy, the rotating speed which were induced by APO in PD rats were significantly decreased after PGXF therapy. In 4 weeks by PGXF treated, there was no significantly increasing of the rotating speed which were induced by APO in PD rats. The abnormal behavioural change were decreased obviously by PGXF therapy; Quantification of Tyrosine Hydroxylase-Positive neurons were increased significantly by PGXF therapy whether with or without LD (P<0.05); (3) PGXF therapy can increase local Glutathione peroxidase and Superoxide dismutase level, and decrease Malonaldehyde and Oxygen free radicals content (P<0.05); (4) PGXF therapy did not affect the expression of DR2 mRNA, but improved the activity of DR2 receptor obviously.Conclusion: The effects of PGXF therapy can decrease the D2 activity, improve the function of Decompensated D2 receptor, remove local Oxygen free radicals, and protect dopamine neurons, and improve Symptoms and development of PD. |
