Objectives: The aim of our study was to reveal the relationship between IGF2BP3and clinico-pathologic features and prognosis of esophageal squamous cell carcinoma(ESCC).Methods: we reviewed the records of 100 patients undergoing complete resection foresophageal squamous cell carcinoma to collect data in T-stage, N-stage, tumordifferentiation and survival. IGF2BP3 protein expression was assessed byimmunohistochemistry in the tissue microarray containing all 100 patients, IGF2BP3mRNA expression was measured by quantitative RT-PCR in another independent setof 33 patients. The mRNA expression level of IGF2BP3 was compared with theprotein expression level in 20 of 33 patients.Results: IGF2BP3 protein expression was observed in 17% (17/100) of primarytumors and 2% (2/100) in normal esophageal mucosa (p=0.000). Compared with lowIGF2BP3 protein expression, increased IGF2BP3 protein expression was associatedwith poor survival of ESCC (5yr survival 45.7%vs23.5%, p=0.032). IGF2BP3 mRNAexpression level in the tumor was higher than that in the normal mucosa in 63.6% of33 patients. No correlation between IGF2BP3 protein expression and mRNAexpression was found in ESCC (p=0.323).Conclusion: Over-expression of IGF2BP3 protein is associated with poor survival ofESCC. IGF2BP3 mRNA is over-expressed in ESCC. No correlation betweenIGF2BP3 protein expression and mRNA expression is found in ESCC. IGF2BP3 maybe involved in the development of ESCC.
