| Fructus Gardeniae is Rubiaceae plant Gardenia jasminoides Ellis' fruit. It a common TCM was used for several thousand years, and it was recorded firstly in Shennong's Herbal, and was regarded as middle grade simultaneously, after that all kinds of books on Chinese herbals recorded it. The grewing area is widespread, such as Hunan, JiangXi, Sichuan, Hupeh, Zhejiang, fukien, GuangDong, Guangxi, Guizhou province and etc. Gardenia is bitter and its property is cold, and it can purg pathogenic fire, reduc fever to induce urination, cool blood to remove pathogenic heat. It has been being used in many diseases, such as hyperpyrexia, jaundice with damp-heat pathogen, jaundice with urine abnorly, bloodystranguria, spitting blood, conjunctival congestion, pyocutaneous disease, contused wound and etc. The processed slices which we studied include Fructus Gardeniae, chao Fructus Gardeniae, jiao Fructus Gardeniae. Fructus Gardeniae' nature is very bitter and cold, so that it can be hurmful to those whose spleen and stomach is not good to cause emesia, but jiao Fructus Gardeniae is not like this. Fructus Gardeniae is good at purging pathogenic fire in triple' warmer, heat-clearing and detoxicating. Jiao Fructus Gardeniae nature is modified and it has hemostasis to cool blood to stop bleeding.We intent to isolate and verify jiao Fructus Gardeniae' chemical compositions on base of Fructus Gardeniae chemical composition isolated systematically, to study the difference of pharmacologic action of chemical compositions between Fructus Gardenia and jiao Fructus Gardeniae. This is significant in theory and practical value.This thesis includes six parts.Part One: Review on the Research Progress of GardeniaConsulted and analyzed the correlative literature data of gardenia from 1960' S to today, including herbal historic evolution, plant resources, chemical compositions, quality control, pharmacological effects, etc.Part Two: Studies on the Chemical Compositions of jiao GareniaIsolated and verified 18 chemical compositions from jiao Gardenia. Four kinds of diterpene pigment, crocetin, crocin-1, crocin-3,and crocin-4．two coumarins, imperatorin and isoimperatorin. two flavanoid, 5-hydroxy-7,3',4',5'-tetramethoxyflavone and 5-hydroxy-6,7,3',4',5'-pentamethoxyflavone. one chromone,2-methyl-3,5-dihydroxychromone one anthraquinone, physcion. three iridoids, geniposide, 6"-O-P-coumaroyl genipingen-tiobioside and genipin-1-β-gentiobioside. one triterpene, ursolic acid. one sterol,β-sitosterol. two porphyrin ring, chlorophyll a and b. one azo, sudanⅢ. Among these composition, eight are isolated for the first time.Part Three: Studies on Fingerprint of Three Kinds of Gardenia SlicesWeight a quantity of Gardenia powder, hyperacoud for a while, methanol as solvent, concentrate, and put the extractum on the HP20 macroporous adsorptive resins chromatographic column, elute with 30%, 50%, 95% alcohol and water, collect the eluting solution quantitively, meter volume, filter, and now the specimen is ok. The result indicates that the change of composition of jiao Fructus Gardeniae' 30% and 50% alcohol parts are not obviously, but 95% alcohol part is better, This is the point of the principle of the uniqe processing.Part Four: Studies on the Main Composition of Three Gardenia SlicesIn this part we had determined contents of the iridoids and pigment compositions, which are the main compositions of Gardenia. The content of iridoids much than that in pigment, these two kinds of compositions changing during of the processing, and pigmeng changed greatlier than iridoids. The pigment quantity in jiao Fructus Gardeniae degressed very extremely, and the composition transformed from crocin-1 to crocin-2, crocin-3 and crocetin, or from crocin-2 to crocin-3 and crocetin and so on.Part Five: Studies on Pharmacological and Pharmacodynamic Action and Pharmacokinetic of Main Alcohol Parts and Crocin-1 and Crocetin.The results indicated that both the main parts of 50%, 95% , crocetin and crocin-1 showded the good effect on antiinflammatory and analgesic effect, but not good on mitigate and choleretic effect. Except for 95% part of stir-baked Gardenia, They all can protect of liver well from injure. Both of the parts of 95% of the two kinds of slices can inhibite the platelet aggregation, and the two chemical compositions can shorten the clotting time extremely.The blood plasma pharmacokinetic results indicate that crocetin Ka 0．83±0．30 (1/h), MRT 3．7±0．44 (h), Cmax 407．06±90．69 (μg/ml), Tmax 1．92±0．20 (h), Vc/f 0．04±0．006 (Mg/μg/ml). The metaboliste from urine and stool are crocetin, the egesting rate is 16．88%, and arrived at the highest top at 12 h and 8 h respectively. crocin-1 Ka 5．474±5．175 (1/h), MRT 5．153±1．072 (h), Cmax 27．55±6．4079(μg/ml), Tmax 2±0 (h), Vc/f 5．0984±3．7089 (Mg/μg/ml). The metaboliste from urine and stool are crocetin, crocin-1, crocin-2, and crocin-3, the egesting rate is 33．33%, and arrived at the highest top at 12 h and 8 h respectively.Part Six: Summarize.Summarize the contents and consults, to elucidate the meaning and new ideas, elucidate the principle of the unique processing of the slice. |
